应用靶向代谢组学技术分析白牛槭叶片不同呈色期类黄酮化合物的差异

摘  要:为深入研究白牛槭(Acer mandshuricum Maxim.)5个呈色期(绿叶期、黄绿叶期、黄叶期、黄红叶期、红叶期)类黄酮化合物积累的变化,利用靶向代谢组学技术分析了白牛槭叶片呈色期类黄酮化合物的差异代谢物种类及质量分数。结果表明:绿叶期、黄绿叶期、黄叶期、黄红叶期、红叶期5个时期叶片中分别检测出36、41、40、48、46种代谢物,囊括了黄酮类及花青素类两大类。质量分数大于1μg·g^(-1)的代谢物包含黄酮类化合物6种、矢车菊素6种、原花青素4种、飞燕草素2种、天竺葵色素1种、芍药花色素1种。随着叶色变红,矢车菊素-3-O-葡萄糖苷质量分数显著升高,其他矢车菊素、天竺葵色素、芍药色素、飞燕草色素也逐渐升高。矢车菊素、天竺葵色素、芍药色素、飞燕草色素在白牛槭叶色变红过程中起到重要作用,其中,矢车菊素是白牛槭叶色变红的决定性因素。To investigate the changes in flavonoid compound accumulation during the five coloration stages(green leaf stage,yellow-green leaf stage,yellow leaf stage,yellow-red leaf stage,red leaf stage)of Acer mandshuricum,targeted metabolomics techniques were employed to analyze the differential metabolites and mass fractions of flavonoid compounds in the leaves.The results showed that 36,41,40,48,and 46 metabolites were detected in the green leaf stage,yellow-green leaf stage,yellow leaf stage,yellow-red leaf stage,and red leaf stage,respectively,encompassing two major categories:flavonoids and anthocyanins.Metabolites with a mass fraction greater than 1μg·g^(-1) included 6 types of flavonoid compounds,6 types of cyanidin,4 types of proanthocyanidin,2 types of delphinidin,1 type of pelargonidin,and 1 type of peonidin.As the leaf color turned red,the mass fraction of cyanidin-3-O-glucoside increased significantly,while the mass fractions of other cyanidins,pelargonidins,peonidins,and delphinidins also gradually increased.Cyanidins,pelargonidins,peonidins,and delphinidins play important roles in the red coloration process of A.mandshuricum leaves,with cyanidin being the decisive factor in the red coloration of A.mandshuricum leaves.
