优化溃结方对LPS诱导人结肠上皮细胞抗炎作用及其作用机制研究

摘　　要：目的观察优化溃结方对脂多糖(lipopolysaccharide,LPS)诱导的人结肠上皮细胞(HCoEpiC)白细胞介素4(interleukin-4,IL-4)、白细胞介素6(interleukin-6,IL-6)的影响。方法根据所给含药血清的不同将经LPS诱导的HCoEpiC分为模型组、SASP组和优化溃结方组,另将未经LPS诱导的HCoEpiC作为正常组。采用QPCR、ELISA法测各组细胞IL-4、IL-6的mRNA及蛋白表达。结果与正常组相比,模型组IL-4的mRNA和蛋白表达减少、IL-6的mRNA和蛋白表达增加(P<0.05);与模型组相比,优化溃结方组和SASP组IL-4表达显著升高(P<0.05),IL-6表达显著降低(P<0.05),且两组间差异无统计学意义(P>0.05)。结论优化溃结方能够增加LPS诱导的HCoEpiC细胞抑炎因子IL-4的表达,减少促炎因子IL-6的表达,为该方的临床应用及后续实验研究提供科学依据。Objective To observe the effects of Youhua Kuijie Decoction(优化溃结方)on interleukin 4(IL-4)and interleukin 6(IL-6)of human colon epithelial cells(HCoEpiC)induced by lipopolysaccharide(LPS).Methods The LPS-induced HCoEpiC was divided into model group,SASP group and Youhua Kuijie Decoction group according to the different drug serum,and the non-LPSinduced HCoEpiC was classified as normal group.The mRNA and protein expression of IL-4 and IL-6 were measured by QPCR and ELISA.Results Compared with normal group,IL-4 mRNA and protein expression decreased and IL-6 mRNA and protein expression increased in model group(P<0.05).Compared with model group,the expression of IL-4 in Youhua Kuijie Decoction group and SASP group was significantly increased(P<0.05),and the expression of IL-6 was significantly decreased(P<0.05),and there was no significant difference between the two groups(P>0.05).Conclusion Youhua Kuijie Decoction can increase the expression of LPS-induced anti-inflammatory factor IL-4 in HCoEpiC cells and reduce the expression of pro-inflammatory factor IL-6,which provides scientific basis for the clinical application and subsequent experimental studies of this prescription.
