补中益气汤对免疫抑制小鼠NK细胞调控作用研究

摘　　要：目的研究补中益气汤及方中中药配伍对环磷酰胺诱导的免疫抑制小鼠脾脏中自然杀伤细胞(NK细胞)调控作用。方法100只小鼠随机分为10组,除空白组外,其余小鼠采用腹腔注射环磷酰胺制备小鼠免疫抑制模型。免疫抑制小鼠随机分为模型组,阳性组,高、中、低剂量中药组,黄芪+白术组,黄芪+党参组,白术+党参组,黄芪+白术+党参组,连续给药17 d后,测量小鼠体质量并计算脾脏及胸腺指数,流式细胞仪检测NK细胞比例,酶标法测试NK细胞活性。结果与空白组比较,模型组小鼠脾脏指数、胸腺指数、NK细胞比例与活性明显降低(P<0.01),表明免疫抑制小鼠模型建立成功。与模型组比较,中药(补中益气汤)组可以有效提高小鼠免疫器官指数、NK细胞比例与活性(P<0.01或P<0.05),其中以中剂量中药组纠正效果最为显著,NK细胞比例高于阳性组,但不及空白组水平,表明方剂可改善免疫抑制状态。黄芪+白术组、黄芪+白术+党参组可提高小鼠免疫器官指数(P<0.01或P<0.05),白术+党参组、黄芪+白术+党参组可提高小鼠NK细胞比例与活性(P<0.01或P<0.05),表明方中主药配伍亦可一定程度提升免疫功能,但效果不及全方组(中剂量中药组)。结论补中益气汤可拮抗环磷酰胺所导致的免疫抑制,增强NK细胞的免疫功能。Objective To investigate the regulatory effect of Buzhong Yiqi Decoction(补中益气汤)and traditional Chinese medicine on natural killer cells(NK cells)in the spleen of cyclophosphamide-induced immunosuppressed mice.Methods A total of 100 mice were randomly divided into 10 groups,except for the blank group,the remaining mice were prepared by intraperitoneal injection of cyclophosphamide to prepare a mouse immunosuppression model.Immunosuppressed mice were randomly divided into model group,positive drug group,high,medium and low dose Chinese medicine group,Huangqi+Baizhu group,Huangqi+Dangshen group,Baizhu+Dangshen group,Huangqi+Baizhu+Dangshen group,after 17 days of continuous administration,the body weight of mice were measured and the spleen and thymus index were calculated,the proportion of NK cells was detected by flow cytometry,and NK cells activity was tested by enzyme tag.Results Compared with the blank group,the spleen index,thymus index,proportion and activity of NK cells in the model group mice were significouldtly reduced(P<0.01),indicating the successful establishment of the immunosuppressive mouse model.Compared with the model group,Chinese medicine(Buzhong Yiqi Decoction)group could effectively improve the immune organ index,NK cells ratio,and activity of mice(P<0.01 or P<0.05),with the medium dose Chinese medicine group showing the most significouldt correction effect.The NK cells ratio was higher than that of the positive group,but lower than that of the blank group,indicating that the formula could improve the immunosuppressive state.Huangqi+Baizhu group and Huangqi+Baizhu+Dangshen group could improve the immune organ index of mice(P<0.01 or P<0.05),while Baizhu+Dangshen group and Huangqi+Baizhu+Dangshen group could increase the proportion and activity of NK cells in mice(P<0.01 or P<0.05),indicating that the main drug combination in the formula could also improve the immune function to a certain extent,but the effect was not as good as whole group(medium dose Chinese medicine group).Conclusio
