辽宁道地药材北五味子的蜜制工艺优化及质量标准评价

摘  要:目的正交试验优选辽宁道地药材北五味子的蜜制工艺,并对蜜五味子炮制品进行质量标准评价。方法研究收集10个不同厂家生产的共10批生北五味子,采用蜜蒸法炮制蜜五味子,以五味子醇甲的含量作为考察指标,采用正交试验优选蜜五味子的最佳炮制工艺;研究对蜜五味子进行性状、水分、总灰分检查;采用薄层色谱法对蜜五味子进行定性鉴别;采用高效液相色谱法(high performance liquid chromatography,HPLC)对蜜五味子中的五味子醇甲进行含量测定。结果正交试验得出蜜五味子的最佳炮制工艺条件:闷润时间1 h,炼蜜量为每100 kg五味子,用10 kg炼蜜,蜜水比例为1∶2,蒸煮时间为40 min。所得10批蜜五味子均性状较佳;该品含水量为12.95%,总灰分为3.36%,均未超过《中华人民共和国药典》标准的16.0%和7.0%;薄层色谱显示10批蜜五味子样品中五味子醇甲均斑点清晰,分离效果好;HPLC色谱检测得出10批蜜五味子样品中五味子醇甲的含量为0.48%~0.70%,平均值为0.59%,高于《中华人民共和国药典》中的含量限度0.40%。结论研究所得北五味子的蜜制工艺稳定可行,建立的质控标准准确度高、专属性强、重复性好,可作为蜜五味子炮制品的质量评价标准。Objective The processing technology of honey Wuweizi(Schisandrae Chinensis Fructus)was optimized by orthogonal test,and its quality standard was evaluated.Methods A total of 10 batches of raw Wuweizi(Schisandrae Chinensis Fructus)from 10 different manufacturers were collected and processed by honey steaming method.With the content of schisandrin A as the index,the best processing technology of honey processing Wuweizi(Schisandrae Chinensis Fructus)was selected by orthogonal test.Thin layer chromatography(TLC)was used to qualitatively identify honey processing Wuweizi(Schisandrae Chinensis Fructus).Thin layer chromatography was used to identify the honey processing Wuweizi(Schisandrae Chinensis Fructus)seed qualitatively.The content of schisandrin A in honey processing Wuweizi(Schisandrae Chinensis Fructus)was determined by high performance liquid chromatography(HPLC).Results The optimum honey processing Wuweizi(Schisandrae Chinensis Fructus)were as follows:dampening time 1 hour,mixing honey 10 kg into 100 kg Wuweizi(Schisandrae Chinensis Fructus),ratio of honey to water 1∶2,cooking time 40 minutes.All the 10 batches of honey processing Wuweizi(Schisandrae Chinensis Fructus)were of good character.The water content of this product was 12.95%and the total ash content was 3.36%,which did not exceed the 16.0%and 7.0%of the Pharmacopoeia of the People's Republic of China standards.TLC showed that the spots of schisandrin A in 10 batches of honey processing Wuweizi(Schisandrae Chinensis Fructus)samples were clear and the separation effect was good.The content of schisandrin A in 10 batches of honey processing Wuweizi(Schisandrae Chinensis Fructus)samples was 0.48%-0.70%,with an average value of 0.59%,which was higher than the limit of 0.40%in Pharmacopoeia of the People's Republic of China.Conclusion The honey production process of Wuweizi(Schisandrae Chinensis Fructus)was stable and feasible,and the established quality control standard had high accuracy,strong specificity and good repeatability,which could be used as
