摘 要:目的观察艾灸是否通过影响氧化应激,维护生殖系统中睾酮之水平,以期为深入研究艾灸延缓生殖衰老机制问题奠定基础。方法以自然衰老小鼠(18月龄)为衰老模型,随机分为衰老组、艾灸组;另设青年组(3月龄)。艾灸组进行麦粒灸双侧足三里穴,艾绒制成麦粒大小的艾炷,置于涂抹凡士林的穴位上,线香引燃,每穴5壮,衰老组只在相同部位做抓取固定动作,不做艾灸,麦粒灸每干预5次间隔2 d。采用HE染色、衰老相关β半乳糖苷酶(senescence-associatedβ-galactosidase,SA-β-gal)染色观察麦粒灸足三里穴干预自然衰老小鼠的效应,并应用ELISA法测量血清睾酮(testosterone,T)含量,WB法测量p53、p21、丙二醛(malondialdehyde,MDA)、锰超氧化物歧化酶(manganese superoxide dismutase,MnSOD)、谷胱甘肽过氧化物酶4(glutathione peroxidase 4,GPX4)表达量。结果HE染色结果显示麦粒灸足三里穴可改善自然衰老小鼠睾丸组织形态学,SA-β-gal染色结果显示麦粒灸足三里穴可改善自然衰老小鼠睾丸衰老程度。与青年组比较,衰老组p53、p21表达均显著升高,表明造模成功,而经过麦粒灸足三里穴干预后,均显著降低;衰老组血清T显著降低,而艾灸组显著上升;在氧化与抗氧化指标方面,衰老组MDA明显上升,而艾灸组MDA水平下降,衰老组MnSOD、GPX4都下降,而艾灸组显著升高。结论麦粒灸足三里穴可以提高自然衰老模型小鼠的血清睾酮水平,而这种作用的发挥与氧化应激密切相关,艾灸通过抑制活性氧(reactive oxygen species,ROS)的产生与增强抗氧化能力,进而阻碍氧化应激,维护了睾丸间质细胞的结构与功能,提高了睾酮水平,从而延缓了睾丸间质细胞的衰老。Objective To observe whether moxibustion affects oxidative stress and maintains the level of testosterone in the reproductive system,so as to lay a foundation for further research on the mechanism of moxibustion in delaying reproductive aging.Methods Natural aging mice(18 months old)were used as aging models and randomly divided into aging group and moxibustion group.A young group(3 months old)was also included.In the moxibustion group,grain-sized moxibustion was applied to bilateral Zusanli(ST36),and moxibustion was made into moxa cones the size of wheat grains and placed on the acupoints coated with vaseline,followed by burning with incense,with 5 cones at each acupoint.In the aging group,only grasping and fixation were performed at the same points without moxibustion,and grain-sized moxibustion was applied 5 times every 2 days.HE staining and senescence-associatedβ-galactosidase(SA-β-gal)staining were used to observe the effect of grain-sized moxibustion at Zusanli(ST36)on natural aging mice.ELISA method was used to measure the content of serum testosterone,and WB method was used to measure the expression of p53,p21,MDA,MnSOD and GPX4.Results HE staining showed that grain-sized moxibustion at Zusanli(ST36)could improve the morphology of testicular tissue in naturally aging mice,while SA-β-gal staining showed that grain-sized moxibustion at Zusanli(ST36)could improve the degree of testicular aging in naturally aging mice.Compared with the youth group,the expression of p53 and p21 in the aging group was significantly increased,indicating successful modeling,while after intervention at the Zusanli(ST36)with grain-sized moxibustion,they were significantly reduced;The serum T level in the aging group significantly decreased,while it significantly increased in the moxibustion group;In terms of oxidation and antioxidant indicators,the aging group showed a significant increase in MDA,while the moxibustion group showed a decrease.The aging group showed a decrease in MnSOD and GPX4,while the moxibustion group showed
