摘  要:建立了一种同时测定拟南芥中6种核苷类成分胸苷、2′-脱氧腺苷、腺苷、2′-脱氧鸟苷、胞苷、鸟苷的高效液相色谱法。拟南芥经前处理后,采用Venusil XBP C18色谱柱(250 mm×4.6 mm×5μm)分离目标化合物,以水-甲醇为流动相进行梯度洗脱,流速为0.5 mL/min,柱温为35℃,在260 nm波长下进行分析。结果表明,6种核苷类成分在0.01~1000μg/mL范围内,线性关系良好(R^(2)≥0.999),检出限在0.10~0.16 mg/kg之间,加标回收率为70.70%~103.09%。本方法操作简单、精密度良好、重复性强且准确度高,能够同时测定拟南芥中6种核苷类成分的含量。A high performance liquid chromatography(HPLC)method was established for the simultaneous determination of 6 nucleosides(thymidine,2′-deoxyadenosine,adenosine,2′-deoxyguanosine,cytidine,guanosine)in Arabidopsis thaliana.After pretreatment of Arabidopsis thaliana,the target compound was separated using a Venusil XBP C18 column(250 mm×4.6 mm×5μm)by gradient elution with water-methanol as the mobile phase.The flow rate was 0.5 mL/min,the column temperature was 35℃,and the analysis was performed at 260 nm wavelength.The results showed that the 6 nucleosides had a good linear relationship(R^(2)≥0.999)within the range of 0.01-1000μg/mL,the detection limit was 0.10-0.16 mg/kg,and the spiked recovery was 70.70%-103.09%.The method has the advantages of simple operation,good precision,strong repeatability and high accuracy.It can simultaneously determine the content of 6 nucleosides in Arabidopsis thaliana.
