基于TLR4/TRIF/STAT1通路探讨大蝉草多糖通过增强免疫抑制黑色素瘤细胞的机制

摘  要:探讨大蝉草多糖(CCPa)通过调节免疫细胞(RAW264.7)活性抑制黑色素瘤细胞(B16F10)的机制。构建RAW264.7和B16F10细胞共培养体系并给予CCPa干预,检测B16F10细胞的增殖能力、凋亡能力和转移能力,明确共培养条件下CCPa对B16F10的抑制作用;另外,采用不同浓度CCPa给药RAW264.7,通过检测RAW264.7的细胞活力、细胞吞噬能力、F-actin蛋白表达、免疫因子水平以及TLR4、TRIF、p-STAT1蛋白的表达,明确CCPa增强RAW264.7免疫活性的具体作用机制。研究结果表明,在共培养体系下,CCPa能协同顺铂抑制B16F10细胞的增殖和转移;RAW264.7细胞经CCPa处理后,细胞的吞噬能力,F-actin蛋白的表达以及免疫因子(IFN-β、IL-6)的水平均明显上升(P<0.05),同时,干预后的细胞内TLR4、TRIF、p-STAT1的表达显著上升(P<0.05)。以上结果表明CCPa可通过TLR4/TRIF/STAT1通路增强RAW264.7细胞免疫活性从而有效抑制B16F10细胞。The specific mechanism of the inhibitory effect of polysaccharide from Cordyceps cicadae(CCPa)on melanoma cells(B16F10)by regulating the activity of immune cells(RAW264.7)was investigated.A co-culture system of RAW264.7 and B16F10 was constructed and intervention of CCPa was given.The inhibitory effect of CCPa on B16F10 was determined by detecting the proliferation,apoptosis and metastasis capacity of B16F10 cells.In addition,different concentrations of CCPa were administered to RAW264.7,and the specific mechanism of the effect of CCPa on enhancing the immune activity of RAW264.7 was elucidated by detecting cellular phagocytic ability,F-actin protein level,immune factor level as well as the expression of TLR4,TRIF and p-STAT proteins of the immune cells.It was found that under the co-culture system,CCPa could inhibit the proliferation and metastasis of B16F10 cells in combination with cisplatin.The phagocytosis ability,expression of F-actin protein and the levels of immune factors(IFN-β,IL-6)of RAW264.7 cells were increased after CCPa treatment(P<0.05).At the same time,the expression of TLR4,TRIF and p-STAT1 were increased significantly(P<0.05).The results reveal that CCPa can effectively inhibit B16F10 cells by enhancing the activity of RAW264.7 through TLR4/TRIF/STAT1 pathway.
