摘 要:目的 研究淫羊藿促进阿尔茨海默病模型小鼠海马神经再生、改善认知功能的作用。方法 将6月龄APP/PS1双转基因小鼠随机分为模型组、淫羊藿组(低、中、高)和阳性对照药组,每日灌胃给药1次,连续28 d。用筑巢与水迷宫实验检测小鼠的生活、学习和记忆能力;用尼氏染色和免疫荧光化学法检测海马尼氏体、成熟神经元、神经干细胞数量及星形胶质细胞活化情况;用透射电镜法检测神经元超微结构;体外培养阿尔茨海默病小鼠海马神经干细胞,检测淫羊藿对其增殖与分化能力的影响。结果 与模型组比较,淫羊藿组小鼠的学习、记忆及生活能力都明显提高;其海马尼氏体与成熟神经元数量明显增加,线粒体损伤明显减轻,突触数量与突触后致密物厚度都明显增加,而活化的星形胶质细胞明显减少,说明淫羊藿抑制了脑内免疫炎性反应,减轻了神经元的损伤;同时还使海马神经干细胞数目明显增多,体外实验也证实淫羊藿能促进阿尔茨海默病神经干细胞增殖,并诱导其向神经元分化。结论 淫羊藿能保护阿尔茨海默病小鼠脑组织减轻神经元损伤,并激活内源神经干细胞促进海马神经再生,有效修复替代其脑内缺损的神经元,从而改善阿尔茨海默病小鼠的认知功能。Objective To study the effect of Epimedium on improving cognitive function via promoting hippocampal neural regeneration in Alzheimer's disease (AD) model mice.Methods 6-month-old APP/PS1 double transgenic mice were randomly divided into model group,Epimedium low,medium and high dose groups and positive control drug group.Mice were administered orally once a day for 28 consecutive days.Using Nesting and Morris water maze experiments to test the daily living,learning and memory abilities of the mice;Nissl staining and immunofluorescence were used to detect the number of Nissl bodies,mature neurons,neural stem cells (NSCs) and the activation of Astrocyte in hippocampus;Transmission electron microscopy was used to detect neuronal ultrastructure.Hippocampal NSCs of AD mice were cultivated in vitro and their proliferation and differentiation were detected.Results Compared with the model group,the learning,memory and living abilities of the Epimedium group mice were significantly improved.The number of Nissl body and mature neuron in the hippocampus were significantly increased.The damage to neuronal mitochondria was significantly reduced,and both the number of synapses and the thickness of postsynaptic dense matter increased significantly,while the number of activated Astrocyte decreased significantly,demonstrating that Epimedium inhibits the immune inflammatory reaction in the brain,and reduced neuronal damage.At the same time,it also significantly increased the number of hippocampal NSCs in the brain.In vitro experiments also confirmed that Epimedium could promote the proliferation of AD-NSCs and induce their differentiation into neurons.Conclusion Epimedium can improve cognitive function in AD mice by protecting brain tissue and activating endogenous NSCs to promote hippocampal neural regeneration.
