摘 要:目的建立安五脂素的高效液相色谱-二极管阵列(HPLC-DAD)含量测定方法,以对护肝片中南五味子掺伪五味子的情况进行研究。方法采用Agilent ZORBAX Eclipse XDB-C18色谱柱(4.6 mm×250 mm,5μm),流动相为甲醇-水(70∶30),柱温30℃,流速1.0 mL/min,检测波长为280 nm,参比波长为360 nm,进样量为10μL。结果安五脂素在浓度在12.064~120.640μg/mL范围内线性关系良好,平均加样回收率为98.80%,RSD为1.07%。结论建立的安五脂素含量测定方法灵敏度高、专属性强、重现性好,可用于护肝片中南五味子掺伪五味子的快速鉴别。Objective HPLC-DAD method for the determination of anwulignan was established to identify the adulterated schisandrae chinensis fructus with schisandrae sphenanthera fructus in hugan tablets.Methods The chromatography was performed on Agilent ZORBAX Eclipse XDB-C18 column(250 mm×4.6 mm,5μm)with mobile phase methanol-water(70:30)at column temperature of 30℃and flow rate of 1.0 mL/min.The detection wavelength was 280 nm,the reference wavelength was 360 nm,and the injection volume was 10μL.Results The linear relationship of anwulignan was good in the concentration range from 12.064 to 120.640μg/mL,the average recovery rate was 98.80%and the RSD value was 1.07%.Conclusion The established method for the determination of anwulignan content has high sensitivity,specificity and reproducibility,and it can be used for the rapid identification of the adulterated schisandrae chinensis fructus with schisandrae sphenanthera fructus in hugan tablets.
