牛膝甾酮对骨肉瘤细胞抑制作用的研究

摘　　要：目的探讨牛膝甾酮对骨肉瘤细胞增殖和凋亡的影响。方法利用浓度0、1、5、10、20 mg/L的牛膝甾酮处理骨肉瘤(MG63、HOS)细胞,通过CCK8实验明确药物对细胞影响及最佳浓度,将最佳药物浓度作为实验组,不加药物作为对照组。通过CCK8、平板克隆和流式细胞仪实验观察牛膝甾酮对细胞增殖及细胞凋亡情况的影响。结果在相同作用时间,不同浓度药物干预后骨肉瘤细胞增殖发生了变化,药物浓度为10 mg/L时,骨肉瘤细胞增殖能力明显下降(P<0.01);同一浓度,随着药物作用时间的延长,细胞增殖能力也会发生变化,而72 h后,细胞增殖能力明显下降(P<0.01)。基于此,后续功能实验采用浓度10 mg/L牛膝甾酮,与对照组进行比较,牛膝甾酮组细胞增殖能力得到明显抑制,而细胞凋亡率明显升高(P<0.01)。与对照组比较,牛膝甾酮组的骨肉瘤细胞集落形成能力明显降低(P<0.01)。结论牛膝甾酮有抑制骨肉瘤细胞增殖及促进细胞凋亡的作用。Objective To investigate the effects of inokosterone on the proliferation and apoptosis of osteosarcoma cells.Methods Osteosarcoma MG63 and HOS cells were treated with the different concentrations of inokosterone(0,1,5,10,20 mg/L),to determine the optimal drug intervention concentration through CCK8,and then the optimal drug concentration was used to intervene in osteosarcoma MG63 and HOS cells as the experimental group,while the control group was not treated with inokosterone.The effects of inokosterone on osteosarcoma cell proliferation and apoptosis were observed through CCK8,plate cloning,and flow cytometry experiments.Results After intervention with different concentrations of the drug at the same time,the proliferation of osteosarcoma cells was changed.When the drug concentration was 10 mg/L,the proliferation ability of osteosarcoma cells significantly decreased(P<0.01).At the same concentration,the proliferation ability of cells was significantly inhibited with the extension of time.After 72 hours,the proliferation of osteosarcoma cell markedly decreased(P<0.01).The subsequent experiments were all intervened with 10 mg/L,and the subsequent experimental indicators were measured.Compared with the control group,the proliferation ability of cells in the experimental group was significantly inhibited,the apoptosis rate of cells in the experimental group significantly increased(P<0.01).Compared with the control group,the ability of osteosarcoma cells to form colonies in the inokosterone group significantly decreased(P<0.01).Conclusion Inokosterone has the effects of inhibiting the proliferation of osteosarcoma cells and promoting cell apoptosis.
