摘 要:为探究山荆子中快速碱性化因子(RALF)调控苹果腐烂病抗性的分子机制,本研究经生物信息学、表达模式分析以及果实瞬时表达,对部分成员调控腐烂病抗性的分子机理进行初步明确。结果表明,山荆子中包含20个RALF成员,进化上可分为6个亚组。顺式作用元件分析结果表明,山荆子RALF可能主要参与生物和非生物逆境信号的响应。在腐烂病侵染后,MbRALF4和MbRALF9响应腐烂病信号且表达量上调。在富士苹果中瞬时表达MbRALF4和MbRALF9降低了苹果果实对腐烂病的抗性。此外,过表达MbRALF4和MbRALF9抑制激发活性氧(ROS)和病原相关分子模式触发的免疫反应(PTI)信号通路基因的表达。综上,山荆子RALF响应生物胁迫,MbRALF4和MbRALF9可负调控腐烂病抗性。本研究结果可为腐烂病抗性研究提供理论基础。To investigate the molecular mechanism of the Malus baccata RALF in regulating Valsa canker resistance and to provide a reference for the prevention and treatment of Valsa canker.The molecular mechanism of members regulating valsa canker resistance was preliminarily clarified based on bioinformatics,expression analysis and fruit transient expression.Results show that there are 20 RALFs in Malus baccata,which can be divided into 6 subgroups.Promoter cis-acting elements analysis showed that the RALF may mainly participate in the response to both biotic and abiotic stress signals.MbRALF4 and MbRALF9 responded to the Valsa canker signal and their expression levels were up-regulated after Vm metabolites(VmM)treatment.Transient expression of MbRALF4 and MbRALF9 in Fuji apple reduced the resistance of apple fruit to Vm.In addition,overexpression of MbRALF4 and MbRALF9 inhibited the expression of genes involved in ROS and PTI signaling pathway.In Conclusion,Malus baccata RALFs responds to biological stress.MbRALF4 and MbRALF9 negatively regulate Valsa canker resistance,which could provide a theoretical basis for the study of Valsa canker resistance.
