基于GC-MS法分析珊瑚七十味丸中挥发油和脂溶性成分

摘　　要：目的:采用气相色谱-质谱(GC-MS)法分析同一厂家10个批次珊瑚七十味丸中的挥发油和脂溶性成分,为后期有效成分研究奠定基础。方法:水蒸气蒸馏法提取珊瑚七十味丸粉末中的挥发油成分,溶剂萃取法提取珊瑚七十味丸粉末中的脂溶性成分,采用GC-MS法分离鉴定,利用NIST14谱库检索对各色谱峰结合有关图谱解析并加以确认,并采用峰面积归一法测定各个化合物的相对百分含量。结果:挥发油成分鉴定出60个化合物,共9类,为烯烃类、芳香烃类、黄酮类、酚类、醚类、羧酸类、酯类、烷烃类、醇类。其中以烯烃类最多,其次为芳香烃类和酯类。脂溶性成分鉴定出42个化合物,共8类,为烯烃类、烷烃类、醇类、芳香烃类、黄酮类、酚类、醚类、羧酸类、酯类。其中以酸类最多,其次为烯烃和醚类。结论:珊瑚七十味丸挥发油主要成分为丁香酚、棕榈酸和(Z)-蒿本内酯等,脂溶性成分为亚油酸、异丁香酚甲醚、肉豆蔻酸等,可能来自于珊瑚七十味丸中的当归、藏党参、藏红花等,有助于进一步明确珊瑚七十味丸的药效物质基础。Objective:Gas chromatography-mass spectrometry(GC-MS)was used to analyze the volatile oil and liposoluble components in 10 batches of Shanhu Qishiwei Wan(SQW)from the same manufacturer,which laid a foundation for the study of effective components in the later research.Methods:Volatile oil was extracted from SQW by steam distillation,liposoluble components were extracted from SQW by solvent extraction.The components was separated and analyzed by GC-MS,identified and confirmed by NIST14 library search.The relative percentage of each compound was determined by peak area normalization method.Results:60 compounds were identified in the volatile oil,belonging to 9 categories,including olefins,aromatic hydrocarbons,flavonoids,phenols,ethers,carboxylic acids,esters,alkanes and alcohols.Among them,olefins were the most,followed by aromatic hydrocarbons and esters.Forty-two compounds were identified from liposoluble components,belonging to 8 categories,including olefins,alkanes,alcohols,aromatic hydrocarbons,flavonoids,phenols,ethers,carboxylic acids and esters.Among them,acids were the most,followed by olefins and ethers.Conclusion:The main components of volatile oil in SQW are eugenol,palmitic acid and(Z)-Artemisia lactone,and the liposoluble components are linoleic acid,isoeugenol methyl ether,myristic acid,etc.,which may come from Danggui(Angelicae Sinensis Radix),Zangdangshen(Codonopsis thalictrifolia Wall.var.mollis(Chipp.)L.T.Shen)and Zanghonghua(Crocus sativus L.)in SQW,which is helpful to further clarify the material basis of efficacy of SQW.
