摘  要:目的:探索美洲大蠊鲜虫提取物对慢性难愈合创面的疗效与机制。方法:用BALB-3T3细胞进行细胞增殖、划痕和transwell实验,免疫印迹法检测细胞上清液中PI3K、Akt、mTOR和VEGF蛋白表达水平;构建大鼠慢性难愈合创面模型,在造模第0、4、8、12、15 d拍照记录创面,计算创面愈合率;每组随机抽取3只取材,用免疫印迹法检测组织中PI3K、Akt、mTOR和VEGF蛋白表达水平。结果:美洲大蠊鲜虫提取物显著促进BALB-3T3细胞的增殖和迁移(P<0.01),且细胞上清液中VEGF蛋白表达量上升(P<0.05);与模型组比较,美洲大蠊鲜虫提取物高、中、低剂量组、康复新液组和急性创面组创面愈合率增加(P<0.05或P<0.01);蛋白免疫印迹结果显示,与慢性模型组比较,美洲大蠊鲜虫提取物胶浆剂高、中、低剂量组、康复新液组和急性创面组PI3K、Akt、mTOR和VEGF蛋白表达量上升(P<0.05或P<0.01)。结论:美洲大蠊鲜虫提取物可促进BALB-3T3细胞的增殖和迁移,并上调PI3K、Akt、mTOR和VEGF蛋白表达来促进慢性难愈合创面的愈合。Objective:To explore the therapeutic effect and mechanism of Periplaneta Americana fresh insect extract on chronic refractory wounds.Methods:BALB-3T3 cells were used for cell proliferation,wound healing and transwell experiments,and the protein expression levels of PI3K,Akt,mTOR and VEGF in the cell supernatant were detected by Western blot.The chronic refractory wound model was established in rats,and the wound healing rate was calculated by photographing the wound on the 0,4,8,12 and 15 days after the modeling.Three samples were randomly selected from each group,and the protein expression levels of PI3K,Akt,mTOR and VEGF were detected by Western blotting.Results:The extracts of fresh Periplaneta Americana significantly promoted the proliferation and migration of BALB-3T3 cells(P<0.01).The expression of VEGF protein in the cell supernatant of Periplanaria Americana extract group was increased(P<0.05).Compared with the chronic model group,the wound healing rate was increased in the high,medium,and low dose groups of Periplanaria Americana fresh insect extract mucils,Kangfuxin solution group,and acute wound group(P<0.05 or P<0.01).The results of Western blot showed that compared with the chronic model group,the protein expressions of PI3K,Akt,mTOR,and VEGF in the high,medium,and low dose groups of Periplanaria Americana fresh insect extract mucine,Kangfuxin solution group,and acute wound group were increased(P<0.05 or P<0.01).Conclusion:The extract of Periplaneta Americana can promote the proliferation and migration of BALB-3T3 cells,and up-regulate the expression of PI3K,Akt,mTOR and VEGF proteins to promote the healing of chronic refractory wounds.
