摘 要:为筛选参与调控籽鹅产蛋性状的关键基因,依据产蛋记录选取300日龄高低产籽鹅各30只,采血并提取基因组DNA,基于基因组重测序结果筛选差异SNPs并注释基因,应用群体分化指数(Fst)选择信号分析后,进行GO功能和KEGG通路富集分析,筛选与籽鹅产蛋性状相关候选基因。结果表明:2~15周高产籽鹅产蛋率显著高于低产籽鹅(P<0.01);重测序共生成数据1556.48 Gb(n=60),平均深度为21.62倍,平均覆盖率为98.78%;高、低产籽鹅分别筛选到12187636和12291795个SNPs,变异主要位于内含子和非编码区;Fst选择信号分析共筛选出829个候选基因,GO功能和KEGG通路富集分析筛选出MAPK9、TGFB2和SGPL1基因与产蛋性能相关。说明MAPK9、TGFB2和SGPL1可作为籽鹅产蛋性状的候选基因。The aim of this study was to screen the key regulatory genes related to egg production traits in Zi geese.Based on the laying records,blood samples of 30 high-production and 30 low-production 300-day-old Zi geese were collected for genomic DNA extraction and whole genome re-sequencing.The whole genome re-sequencing results were used to screen differential SNPs and annotate genes.After performing the Fixation index(Fst)for signal analysis,GO function and KEGG pathway enrichment analyses were conducted to screen for candidate genes associated with the egg-laying traits of geese.The laying rate of high-production Zi geese was significantly higher than that of low-production during 2 to 15 weeks(P<0.01).Whole genome re-sequencing yielded 1556.48 Gb(n=60)data with an average depth of 21.62,and the the average coverage rate was 98.78%.High and low production Zi geese were screened for 12187636 and 12291795 SNPs,respectively,with the variations mainly located in introns and non-coding regions.A total of 829 candidate genes were identified by Fst selection signature,and MAPK9,TGFB2 and SGPL1 genes were associated with egg production trait by GO function and KEGG pathway enrichment analysis.The results indicated that the three selected candidate genes had influence on laying traits in Zi geese.
