摘 要:精子顶体反应研究是受精生物学最重要的基础研究领域之一。马鹿是目前我国饲养的最主要的特种经济动物之一,但目前有关马鹿体外受精生物学机理的相关研究,国内外的报道均较少。试验旨在运用上浮法使马鹿精子获能,利用钾离子载体缬氨霉素(valinomycin,Val)诱发精子顶体反应,探究其对顶体反应的影响、最佳反应体系及离子浓度变化,为提高鹿科动物精子的体外培养及体外受精效率提供参考,同时为鹿科动物受精机理的探究提供基础数据。结果表明:0.01~50.00μmol/L Val均能提高马鹿精子顶体反应发生率,其中最佳反应体系为0.50μmol/L Val处理5.0 min,顶体反应发生率可达到(74.48±0.79)%;Val诱导未获能马鹿精子的顶体反应率为(33.44±3.47)%,且与其诱导已获能马鹿精子的顶体反应率之间差异显著(P<0.05);Val在诱导顶体反应时能改变培养液中Ca^(2+)、K^(+)浓度。说明Val能促进马鹿精子的顶体反应,并具有离子转运调节作用。The study of the sperm acrosome reaction is one of the most fundamental fields in fertilization biology.At present,the wapiti(Cervus elaphus)is one of the economically specialized animals raised in China.However,the exploration of the biological mechanisms of in vitro fertilization in wapiti are less reported both domestically and internationally.The aim of this study was to investigate the impact of valinomycin(Val)on the acrosome reaction,the optimal reaction system and ion concentration changes through capacitating wapiti sperm by using the float-up method and inducing the sperm acrosome reaction with Val.This research intended to provide scientific reference for improving the in vitro culture and in vitro fertilization efficiency of sperm and offer foundational data for the exploration of the fertilization mechanisms in cervids.The results suggested that Val at concentrations of 0.01 to 50.00μmol/L could enhance the acrosome reaction rate of wapiti sperm.The optimal reaction system was to treat wapiti sperm with 0.50μmol/L Val for 5.0 min,which achieved an acrosome reaction rate of(74.48±0.79)%,showing a significant difference from that of(33.44±3.47)%of Val-induced non-capacitated wapiti sperm(P<0.05).Moreover,the concentrations of Ca^(2+)and K^(+)in the culture medium varied during the acrosome reaction induced by Val.This study implied that Val could promote the sperm acrosome reaction in wapiti and possess the function of ion transport regulation.
