摘 要:选择55头黟县黑猪的血液样品,采用PCR(聚合酶链式反应)和PAGE(聚丙烯酰胺凝胶电泳)法对14个微卫星标记进行分型,分析黟县黑猪群体的遗传多样性。结果表明:在黟县黑猪群体中,14个微卫星位点共检测出171个等位基因,平均每个位点约12个,有效等位基因数为3.11-12.37,观测杂合度范围为0.31-1;14个微卫星位点多态性含量为0.65-0.91,均具有高度多态性,累积排除概率达99.9%,说明选择的14个微卫星标记能够对黟县黑猪的亲缘关系进行鉴定。The blood samples of 55 Yixian black pigs were selected in this experiment,and 14 microsatellite markers were typed by PCR(Polymerase chain reaction)and PAGE(Polyacrylamide gel electrophoresis)to analyze the genetic diversity of Yixian black pig population.The results showed that a total of 171 alleles were detected at 14 microsatellite loci in Yixian black pig population,with an average of about 12 alleles per locus.The number of effective alleles was 3.1112.37,and the observed heterozygosity ranged from 0.31 to 1.The polymorphism content of 14 microsatellite loci was 0.65—0.91,all of which were highly polymorphic,and the cumulative exclusion probability was 99.9%,which indicated that the selected 14 microsatellite markers could identify the genetic relationship of Yixian black pigs.
