摘 要:目的研究鱼酱排毒鼻喷雾剂对急性鼻窦炎大鼠的治疗作用。方法采用单侧鼻腔填塞浸有金黄色葡萄球菌菌液的膨胀海绵建立大鼠急性鼻窦炎模型。根据急性鼻窦炎症状评分表评估造模结果。将造模成功的大鼠分为正常组、模型组、中药组和阳性药物(糠酸莫米松鼻喷雾剂)组。收集各组大鼠鼻腔灌洗液进行细菌培养;扫描电镜观察各组大鼠鼻纤毛损伤情况;HE染色检测各组大鼠鼻黏膜病损情况;酶联免疫吸附试验(ELISA)检测各组大鼠血清中TNF-α、IL-6、IL-1β水平;实时荧光定量聚合酶链式反应(RT-PCR)以及蛋白免疫印迹法(Western Blot)检测各组大鼠鼻黏膜组织中TNF-α、IL-6、IL-1β蛋白表达情况。结果与模型组相比,鱼酱排毒鼻喷雾剂组大鼠鼻腔内细菌生长受到抑制;鼻纤毛及鼻黏膜损伤明显减轻;血清及鼻黏膜组织中TNF-α、IL-6、IL-1β水平明显降低(P<0.01)。结论鱼酱排毒鼻喷雾剂在抑制急性鼻窦炎大鼠鼻腔细菌生长繁殖、改善大鼠鼻纤毛损伤及体内炎症因子的分泌效果优于糠酸莫米松鼻喷雾剂,可以作为急性鼻窦炎的治疗用药。Objective To study the therapeutic effect of Yujiang Paidu Nasal Spray on acute sinusitis in rats.Methods The rat model of acute sinusitis was established by unilateral nasal cavity filling with an expansive sponge impregnated with Staphylococcus aureus solution.The modeling results were evaluated according to the acute sinusitis rating scale.The rats were divided into normal group,model group,Yujiang Paidu Nasal Spray group and positive drug(Mometasone Furoate Nasal Spray)group.The nasal lavage fluid of each group was collected for bacterial culture.The damage of nasal cilia in each group was observed by scanning electron microscope.The nasal mucosal lesions were detected by HE staining.The serum levels of TNF-α,IL-6 and IL-1β were detected by enzyme-linked immunosorbent assay(ELISA).Real-time fluorescence quantitative polymerase chain reaction(RT-PCR)and Western Blot were used to detect the expression of TNF-α,IL-6 and IL-1β in nasal mucosa of rats in each group.Results Compared with the model group,the growth of bacteria in the nasal cavity of rats in Yujiang Paidu Nasal Spray group was inhibited.The injury of nasal cilia and mucosa was significantly reduced.The levels of TNF-α,IL-6 and IL-1β in serum and nasal mucosa were significantly decreased(P<0.01).Conclusion Yujiang Paidu Nasal Spray is better than Mometasone furoate nasal spray in inhibiting the growth and reproduction of nasal bacteria,improving the damage of nasal cilia and the secretion of inflammatory factors in rats with acute sinusitis,and can be used as a treatment for acute sinusitis.
