摘 要:目的:探讨乳核内消液对于子宫肌瘤大鼠的药效学作用及其机制。方法:采用雌激素加孕激素负荷法建立子宫肌瘤大鼠模型,将造模成功的大鼠随机分为模型组、米非司酮组、桂枝茯苓组和乳核内消液高(4.0m L/kg)、中(2.0 mL/kg)、低(1.0 mL/kg)剂量组。各给药组给予相应药物干预治疗,空白组和模型组给予等体积的纯化水。60 d后,获取子宫标本,测定子宫脏器系数、平滑肌厚度指标;HE染色进行子宫组织病理学观察;ELISA检测血清雌二醇(E2)、孕激素(P)表达水平;Western blotting检测子宫组织中Wnt和β-catenin表达水平。结果:乳核内消液高、低剂量可明显降低子宫肌瘤模型大鼠子宫脏器系数,与模型组比较,差异有统计学意义(P<0.05);乳核内消液能明显改善子宫平滑肌层增厚(P<0.01)、平滑肌排列紊乱、平滑肌纤维增生肥大等病理改变;乳核内消液高、中、低剂量能明显降低模型大鼠血清E2、P水平,与模型组比较,差异均有统计学意义(P<0.01);乳核内消液高、中、低剂量能明显降低模型大鼠子宫组织中Wnt-5b蛋白表达,与模型组比较,差异均有统计学意义(P<0.05);乳核内消液高、低剂量能明显降低模型大鼠子宫组织中β-Catenin蛋白表达,与模型组比较,差异均有统计学意义(P<0.01)。结论:乳核内消液能够显著降低子宫肌瘤大鼠子宫脏器系数及性激素水平,改善大鼠子宫组织病理学变化,其机制可能与调节Wnt/β-catenin信号通路有关。Objective:To investigate the pharmacodynamic effect and mechanism of Ruhe Neixiao Liquid on hysteromyoma rats.Methods:The rat model of uterine leiomyoma was established by estrogen and progesterone loading method.The rats with successful modeling were randomly divided into model group,mifepristone group,Guizhi Fuling(桂枝茯苓)group,Ruhe Neixiao Liquid high dose(4.0 mL/kg)group,Ruhe Neixiao Liquid medium dose(2.0 mL/kg)group and Ruhe Neixiao Liquid low dose(1.0 mL/kg)group.Each group was given corresponding drugs,while the blank group and model group were given equal volume of purified water.After 60 days,the uterus specimens were obtained,and the uterine organ coefficient and smooth muscle thickness index were measured.HE staining was used for pathological observation of uterine tissue.The levels of serum estradiol(E2)and progesterone(P)were detected by ELISA.The expression levels of Wnt andβ-catenin in uterine tissues were detected by Western blotting.Results:The high and low doses of Ruhe Neixiao Liquid could significantly reduce the uterine organ coefficient of uterine fibroids model rats,and compared with the model group,the difference was statistically significant(P<0.05).Ruhe Neixiao Liquid can significantly improve the pathological changes of uterine smooth muscle layer thickening(P<0.01),smooth muscle arrangement disorder,smooth muscle fiber hyperplasia and hypertrophy.The high,medium and low doses of Ruhe Neixiao Liquid could significantly reduce the levels of serum E2 and P in model rats,and the difference was statistically significant compared with the model group (P<0.01). The high, medium and low doses of Ruhe Neixiao Liquid could significantly reduce the expression of Wnt-5b protein in uterine tissue of model rats, and the difference was statistically significant compared with the model group (P<0.05).
