摘 要:目的:探讨淫羊藿、当归及其配伍通过TGF-β1/Smads信号通路防治绝经后骨质疏松症的作用机制。方法:选取42只雌性未育SD大鼠随机分假手术组(7只)和联合造模组(35只),采用双侧卵巢摘除联合地塞米松肌内注射法建立病证结合的绝经后骨质疏松症模型;造模6周后,将造模成功的30只大鼠随机分为模型组、淫羊藿组、当归组、配伍组、戊酸雌二醇组,每组6只,并给予相应药物治疗8周。治疗结束后,采用苏木素-伊红(HE)染色法观察股骨组织形态,ELISA法检测大鼠血清TGF-β1水平,免疫荧光法检测骨组织中TGF-β1、Smad2、Smad3、Smad7蛋白表达,实时荧光定量PCR法检测TGF-β1 mRNA、Smad2 mRNA、Smad3 mRNA和Smad7 mRNA表达。结果:模型组大鼠TGF-β1、Smad2/3表达水平均低于假手术组(P<0.01),Smad7的表达水平高于假手术组(P<0.01);干预治疗后,配伍用药能显著上调血清TGF-β1的表达水平(P<0.01);各给药组均能显著上调股骨组织中TGF-β1、Smad2/3蛋白及Smad2/3 mRNA表达水平(P<0.01),下调股骨组织中Smad7蛋白及Smad7 mRNA表达水平(P<0.05),且与单独用药相比,配伍用药的效果更加明显(P<0.01)。结论:淫羊藿、当归及其配伍有改善骨代谢、修复骨显微结构的作用,其作用机制可能为通过调控TGF-β1/Smads信号通路,改善相关细胞因子的异常表达,从而发挥骨保护作用以防治骨质疏松症。Objective:To investigate the effect of Yinyanghuo(Epimedium),Dangui(Angelica sinensis)and their combination on the prevention and treatment of postmenopausal osteoporosis(PMOP)through TGF-β1/Smads signaling pathway.Methods:A total of 42 female infertile rats were divided into sham operation group(n=7)and combined model group(n=35).Disease-associated postmenopausal osteoporosis rat model was established by bilateral ovariectomy combined with intramuscular injection of dexamethasone.After 6 weeks of modeling,30 combined model group rats were randomly divided into model group,Yinyanghuo(Epimedium)group(Epimedium group),Dangui(Angelica sinensis)group(Angelica sinensis group),Yinyanghuo(Epimedium)and Dangui(Angelica sinensis)combination group(combination group),and estradiol valerate group,6 rats in each group.Each group was given corresponding medication treatment for 8 weeks.At the end of treatment,the morphology of bone tissue was observed by HE staining. The serum level of TGF-β1 was detected by ELISA. The protein expressions of TGF-β1, Smad2, Smad3 and Smad7 were detected by immunofluorescence and the expressions of TGF-β1 mRNA, Smad2 mRNA, Smad 3 mRNA and Smad7 mRNA were detected by qRT-PCR. Result: The model group showed lower expression levels of TGF-β1 and Smad2/3 than sham operation group (P<0.01), while higher expression level of Smad7 than sham operation group (P<0.01). After intervention, the expression level of TGF-β1 in serum was significantly up-regulated by the combination of the two drugs (P<0.01). The protein expression levels of TGF-β1 and Smad2/3 and expression levels of Smad2/3 mRNA in the femoral tissue were significantly up-regulated in each administration group (P<0.01), while the protein expression level of Smad7 and expression level of Smad7 mRNA in the femoral tissue were significantly down-regulated in each administration group (P<0.05).
