复方钩藤降压片(天苓降压片)对自发性高血压大鼠P2Y6受体及炎症因子水平的影响

摘　　要：目的以SHR和WKY大鼠为实验对象,通过测量相关指标,观察复方钩藤降压片对P2Y6受体的表达及炎症水平的影响。方法60只SHR随机分为5组,即模型对照组、缬沙坦组、阻断剂组、钩藤组、中西药组,另12只WKY大鼠设置为空白对照组,按相应的处理方法连续干预8周后取材,测算HMI和LVMI、HE染色观察左室心肌形态、免疫组化法检测心脏IL-1β、IL-6和TNF-α的蛋白表达、RT-PCR法测定IL-6、TNF-α、IL-1β、P2Y6 mRNA的表达、Western-Blotting法测定心肌IL-6、TNF-α、IL-1β、P2Y6的蛋白表达。结果⑴自发性高血压大鼠心室重量及左室质量较正常大鼠明显增加,是良好的高血压LVH的研究模型;⑵自发性高血压大鼠炎症因子IL-6、TNF-α、IL-1β血清浓度增加,相应的mRNA及蛋白表达增强,提示自发性高血压大鼠炎症活动增强;⑶阻断P2Y6受体,IL-6、TNF-α、IL-1β血清浓度、mRNA及蛋白表达均明显降低,提示P2Y6受体可能是自发性高血压大鼠炎症反应的关键位点;⑷经复方钩藤降压片治疗后,IL-6、TNF-α、IL-1β血清浓度、mRNA及蛋白表达均降低,提示复方钩藤降压片一定程度上能阻断P2Y6受体的表达,且与西药缬沙坦表现出协同作用。结论复方钩藤降压片一定程度上能阻断P2Y6受体的表达,从而降低SHR的慢性炎症水平。Objective To observe the effect of Compound Uncaria Jiangya Tablet on the expression of P2Y6 receptor and inflammation levels in SHR by measuring relevant indicators.Methods 60 SHRs were randomly divided into 5 groups,namely model control group,valsartan group,blocker group,uncaria hook group,and traditional Chinese and Western medicine group.The other 12 WKY rats were set as blank control group.After 8 weeks of continuous intervention according to the corresponding treatment methods,HMI and LVMI were measured,HE staining was used to observe left ventricular myocardial morphology,and immunohistochemistry was used to detect the protein expression of IL-1β,IL-6 in the heart,RT-PCR determination the mRNA expression of IL-6,TNF-α,IL-1β,P2Y6,Western Blotting assaying the protein expression of P2Y6,IL-6,TNF-α,IL-1β.Results 1.Spontaneous hypertensive rats have significantly increased ventricular weight and left ventricular mass compared to normal rats,making them as a good research model for hypertensive LVH.2.Inflammatory factors IL-6,TNF-α,IL-1β increased in spontaneously hypertensive rats,indicating an increase in inflammatory activity in spontaneously hypertensive rats.3.Blocking P2Y6 receptors,the serum concentration of IL-6,TNF-α and IL-1β,as well as the mRNA and protein expression were significantly reduced too,indicating that P2Y6 receptor may be a key site of inflammatory response in spontaneously hypertensive rats.4.After treatment with Compound Uncaria Hypotension Tablets,the serum concentration of IL-6,TNF-α,IL-1βdecreased,as well as the mRNA and protein expression,suggesting that Compound Uncaria Jiangya Tablets can to some extent block the expression of P2Y6 receptors and exhibit synergistic effects with the Valsartan.Conclusion Compound Gouteng Jiangya tablets can extent block the expression of P2Y6 receptors,thereby reducing the level of chronic inflammation in SHR.
