摘 要:目的优选凤仙透骨草中东莨菪内酯的最佳提取工艺和纯化工艺并对其吸附机制进行研究。方法采用HPLC法测定东莨菪内酯的含量,基于正交试验设计法,考察提取温度、乙醇浓度、提取时间三个因素对东莨菪内酯含量的影响。对大孔树脂种类进行筛选,优化东莨菪内酯的富集纯化工艺,并分析其吸附机制。结果超声提取凤仙透骨草中东莨菪内酯的最佳工艺为提取温度60℃,乙醇浓度为80%,提取时间30 min;HPD-100大孔树脂对东莨菪内酯具有较好的纯化能力,吸附动力学符合准二级动力学模型,等温吸附数据符合Freundich模型。最佳纯化工艺为上样量2 BV,上样流速4 BV/h,水洗脱用量3 BV,洗脱液乙醇浓度60%,洗脱流速4 BV/h,洗脱体积6 BV。此工艺得到的东莨菪内酯转移率为73.46%,纯度提高了7.64倍。结论优选出的超声提取和大孔树脂纯化东莨菪内酯的工艺稳定可靠。Objective To select the best extraction process of scopoletin and the purification process of macroporous resin and study its adsorption mechanism.Methods The content of scopoletin was determined by HPLC and the influence of extraction temperature,ethanol concentration and time based on the orthogonal test design method.The types of macroporous resin were screened to optimize the enrichment and purification process and analyze the adsorption mechanism.Results The optimal extraction process of ultrasound was 60℃,ethanol concentration of 80% and extraction time of 30 min;HPD-100 macroporous resin has good purification ability for scopoletin,the adsorption dynamics meets the quasi-secondary kinetic model and the isothermal adsorption data meet the Freundich model.The best purification process is 60% of 4 BV/h,of 4 BV/h,water eluting volume and 6 BV.The transfer rate was 73.46% and the purity increased 7.64 times.Conclusion The preferred ultrasonic extraction process and the purification process of macroporous resin are stable and reliable.
