摘 要:药物容偏性是药物即使漏服一剂或多剂,依然能维持药效的性质。容偏性与药物的药动学和药效学特征有关。一般而言,半衰期长或药效持续时间长的药物有较高的容偏性。容偏性较高的药物可降低不依从行为对治疗效果的影响,提高治疗成功的概率。该文比较了容偏性和依从性,并从药动学和药效学的角度,对容偏性现有评价方法进行综述,总结容偏性的研究方法及在药物全生命周期中的应用,为新药研发和临床合理用药提供参考。Forgiveness is a property of a drug maintaining its therapeutic efficacy even if one or more doses are missed.It is an inherent function of a drug’s pharmacokinetic and pharmacodynamic properties.Generally,a drug may be forgiving if it has long half-life or duration of action.Non-adherence is a common event in real-world settings.Therefore choosing a drug with higher for⁃giveness can greatly boost the probability of successful attaining treatment targets.This review introduced the concept of forgive⁃ness and highlighted its difference with adherence.After reviewing existing evaluation metrics of forgiveness based largely upon pharmacokinetics and pharmacodynamics,a summary of research methods was provided with advantages and disadvantages of each study.Lastly practical examples of how forgiveness is applied in the realm of drug development,formulation modification and therapeutic regimen optimization were summarized for guiding future researches on developing new drugs and rational drug dosing.
