补肾促排卵汤促进骨骼肌葡萄糖转运体GLUT4膜转位改善PCOS大鼠胰岛素抵抗的作用研究

摘  要:目的:探讨补肾促排卵汤改善多囊卵巢综合征(PCOS)大鼠胰岛素抵抗是否与促进骨骼肌细胞葡萄糖转运体(GLUT)4膜转位有关。方法:24只雌性SD大鼠随机分为正常对照组(8只)和PCOS模型组(16只),PCOS模型组连续21 d灌服来曲唑,将造模成功的16只大鼠再随机分为模型组、补肾促排卵汤组,每组8只。补肾促排卵汤组连续14 d灌服补肾促排卵汤。干预结束后,空腹测量各组大鼠体质量,ELISA检测血清睾酮、雌二醇水平及空腹胰岛素水平,生化检测空腹血糖水平,免疫组织化学检测比目鱼肌GLUT4的表达情况,Western blot技术测定比目鱼肌GLUT4、AKT2、p-AKT2、AS160、p-AS160蛋白表达水平及比目鱼肌质膜GLUT4蛋白表达水平。结果:与正常对照组相比,模型组大鼠体质量、空腹血糖、空腹胰岛素以及血清睾酮水平显著升高(P<0.05),雌二醇水平显著降低(P<0.05),大鼠卵巢呈多囊样改变,比目鱼肌质膜GLUT4及比目鱼肌p-AKT2、p-AS160蛋白含量显著降低(P<0.05)。与模型组相比,补肾促排卵汤组大鼠空腹血糖、空腹胰岛素水平显著降低(P<0.05),血清睾酮水平显著降低,雌二醇水平升高(P<0.05),有效改善卵巢形态,比目鱼肌质膜GLUT4蛋白及比目鱼肌p-AKT2、p-AS160蛋白显著上升(P<0.05)。结论:补肾促排卵汤改善PCOS大鼠胰岛素抵抗,其机制可能与激活AKT2/AS160信号通路,促进骨骼肌GLUT4蛋白转位有关。Objective:To investigate whether the improvement of insulin resistance in rats with polycystic ovary syndrome(PCOS)by Bushen Cupailuan decoction(BCD)is related to the promotion of glucose transporter GLUT4 membrane translocation in skeletal muscle cells.Methods:Twenty-four female SD rats were randomly divided into the control group(n=8 rats)and the PCOS model group(n=16 rats).The PCOS model group was administered letrozole for 21 consecutive days,and the 16 rats that had been successfully modeled were randomly divided into the model group and the BCD group,with 8 rats in each group.BCD was administered for 14 consecutive days in BCD group.After the intervention,the body mass of each group was measured on empty stomach,the levels of serum testosterone,estradiol and fasting insulin were detected by ELISA,the levels of fasting blood glucose were detected by biochemistry,and the expression of GLUT4 in soleus muscle was detected by immunohistochemistry.Western blot assay was used to determine the expression levels of GLUT4,AKT2,p-AKT2,AS160,p-AS160 in soleus muscle and GLUT4 in soleus muscle membrane.Results:Compared with the control group,rats in the PCOS model group had significantly higher level of body weight,the serum of fasting blood-glucose,fasting insulin and testosterone(P<0.05);and the estradiol was lower(P<0.05).The ovaries showed polycystic change in the model group.The level of cytoplasmic membrane GLUT4 and the protein levels of p-AKT2,p-AS160 in muscle soleus remarkably decreased in the model group(P<0.05).Compared with the PCOS model group,the levels of fasting blood-glucose and fasting insulin were significantly reduced(P<0.05),the serum levels of testosterone were significantly reduced,and the estradiol levels increased(P<0.05)in BCD group.BCD can effectively improve ovarian morphology,increase the level of cytoplasmic membrane GLUT4 and the protein levels of p-AKT2,p-AS160 in muscle soleus(P<0.05).Conclusion:Bushen Cupailuan decoction improves insulin resistance in PCOS rats,and its mechanism may be
