摘 要:目的建立一种测定胆石通胶囊中咖啡酸、柴胡皂苷A、夏佛塔苷和绿原酸含量的RP-HPLC法。方法在色谱柱Waters spherisorb ODS2 C_(18)(250 mm×4.6 mm,5μm),柱流量1.0 mL·min^(-1);柱温30℃;以乙腈(流动相A)-0.3%磷酸溶液(流动相B)为流动相进行梯度洗脱,咖啡酸、柴胡皂苷A、夏佛塔苷和绿原酸的检测波长分别为323、210、272和327nm,进样量为10μL的色谱条件下建立测定咖啡酸、柴胡皂苷A、夏佛塔苷和绿原酸成分含量的RP-HPLC法。结果咖啡酸、柴胡皂苷A、夏佛塔苷和绿原酸分别在质量浓度0.02991~0.2991、0.4740~4.7400、0.07118~0.7118和0.3852~3.8520μg·mL^(-1)内具有良好的线性,平均回收率分别为100.18%、99.52%、100.20%和100.23%;仪器精密度和方法重复性良好(RSD均在5.0%以内)。结论该RP-HPLC法具有操作简便、快速和高效等优点,可用于胆石通胶囊中咖啡酸、柴胡皂苷A、夏佛塔苷和绿原酸含量的测定。AIM To establish a reverse-phase high performance liquid chromatography(RP-HPLC)method for determination of caffeic acid,saikosaponin A,schaftoside and chlorogenic acid in Danshitong capsules.METHODS A RPHPLC method was developed under the following conditions:A Waters spherisorb ODS2 C_(18) column(250 mm×4.6 mm,5μm)was employed with a flow rate of 1.0 mL·min^(-1)and a column temperature of 30℃.Gradient elution was carried out using acetonitrile(mobile phase A)and a 0.3%phosphoric acid solution(mobile phase B).Detection wavelengths for caffeic acid,saikosaponin A,schaftoside and chlorogenic acid were set at 323,210,272 and 327 nm,respectively.The injection volume was 10μL for quantification of caffeic acid,saikosaponin A,schaftoside,and chlorogenic acid contents.RESULTS Caffeic acid,saikosaponin A,schaftoside and chlorogenic acid showed good linear relationships in concentration range of 0.02991-0.2991,0.4740-4.7400,0.07118-0.7118 and 0.3852-3.8520μg·mL^(-1).Average recoveries were 100.18%,99.52%,100.20%and 100.23%,respectively.Both instrumental precision and method reproducibility were within 5.0%.CONCLUSION The method is characterized by its ease of operation,rapidity,and high efficiency,making it suitable for the determination of caffeic acid,saikosaponin A,schaftoside and chlorogenic acid content in the Danshitong capsule.
