运动训练对2型糖尿病小鼠生化指标及肾损害的影响机制研究

摘  要:目的了解运动训练对2型糖尿病(T2DM)小鼠的体质量、血糖水平、糖化血红蛋白(HbA1c)、尿微量白蛋白等的影响,探讨运动训练基于转化生长因子-β(TGF-β)/Smad信号传导对因糖尿病所致肾损害的影响机制。方法将60只T2DM小鼠随机分为实验组与对照组(各30只),实验组小鼠进行为期8周的运动训练(10 m·min^(-1),每天60 min,每周5天),对照组小鼠久坐不动。每4周检测两组小鼠体质量、血糖水平及上述生化指标。运动训练结束后将所有小鼠处死并检测其肾脏组织TGF-β1、Smad3、Smad7蛋白表达,并进行苏木精-伊红染色和Masson染色观察肾脏组织变化。结果运动训练后实验组小鼠体质量、HbA1c、尿微量白蛋白、肝性脂肪酸结合蛋白水平均低于对照组(P<0.01),空腹血糖、进食后血糖水平两组间比较无明显差异(P>0.05)。实验组小鼠TGF-β1、Smad3蛋白表达低于对照组,Smad7蛋白表达高于对照组(P<0.01)。结论运动训练能控制T2DM小鼠的体质量,对保护肝肾功能有积极影响,有利于减轻T2DM所致肾损害。Objective To investigate the effect of exercise training on the body mass,blood glucose level,glycosylated hemoglobin(HbA1c),microalbuminuria(MAU)of rats with type 2 diabetes mellitus(T2DM)a nd ex plore the mechanism of exercise training effect based on TGF-β/Sm ad sig n a l i ng pathway on renal damage caused by T2DM.Methods Sixty T2DM rats were randomly divided into experimental and control group with 30 in each,rats in experimental group received 8 week exercise training(10 meters·min^(-1),60 minutes/day,5 days/week),controls were sedentary.Body mass,blood glucose level and biochemical indicators for both groups were detected every 4 weeks.After exercise training all rats were sacrificed,the protein expressions of TGF-β1,Smad3 and Smad7 in kidney tissue detected,a nd hematoxylin a nd eosin(HE)a nd Masson's trich rome(Masson)staining perfor med to observe tissue changes.Results After exercise training body mass as well as HbA1c,MAU and hepatic fatty acid binding protein level were lower in experimental than control group(P<0.01),intergroup differences in fasting blood glucose and blood glucose level after eating weren't significant(P>0.05).The protein expressions of TGF-β1 and Smad3 were lower,that of Smad7 washigher in experimental than control group(P<0.01).Conclusion Exercise training can control body mass of T2DM rats,protect hepatic and renal functions,and it is beneficial to alleviate renal damage caused by the T2DM.
