摘  要:目的通过构建慢性鼻⁃鼻窦炎上皮细胞损伤模型,探索慢性鼻⁃鼻窦炎上皮细胞损伤中炎症与AMPK/mTOR信号通路的关系。方法采用不同浓度TNF⁃α刺激鼻粘膜上皮细胞诱导慢性鼻⁃鼻窦炎的鼻粘膜上皮细胞模型。采用Elisa检测正常鼻粘膜上皮细胞(正常组)和慢性鼻⁃鼻窦炎的鼻粘膜上皮细胞(模型组)中炎症因子水平,采用TUNEL检测正常组和模型组的鼻粘膜上皮细胞凋亡情况,采用WB检测正常组和模型组在AMPK/mTOR信号通路相关蛋白(AMPK,p⁃AMPK,mTOR,p⁃mTOR)的相对表达量。结果较0 ng/mL、1ng/mL TNF⁃α处理的慢性鼻⁃鼻窦炎的鼻粘膜上皮细胞,p⁃AMPK蛋白在10 ng/mL TNF⁃α处理组中上调,差异有统计学意义(P<0.05),而AMPK蛋白组间比较差异无统计学意义(P>0.05)。较正常组,模型组中的TNF⁃α与IL⁃1β水平升高,细胞凋亡率降低,p⁃AMPK/AMPK蛋白表达含量上升,p⁃mTOR/mTOR蛋白表达含量显著下降,差异均有统计学意义(P<0.05)。结论TNF⁃α通过调控AMPK/mTOR通路诱导慢性鼻⁃鼻窦炎的鼻粘膜上皮细胞损伤。Objective The relationship between inflammation and AMPK/mTOR signaling pathway in chronic nasosinusitis was explored by constructing the epithelial cell injury model of chronic nasosinusitis.Methods Nasal mucosal epithelial cells were stimulated with different concentrations of TNF⁃αto induce rhinitis cell model The content of inflammatory factors in normal nasopharyngeal epithelial cells(normal group)and rhinitis cells(model group)was detected by Elisa,the apoptosis of nasopharyngeal epithelial cells in normal group and model group was detected by TUNEL,and the AMPK/mTOR signaling pathway related proteins(AMPK,p⁃AMPK,mTOR,MTOR)in normal group and model group were detected by WB.The relative expression of p⁃mTOR.Results Compared with 0 ng/mL and 1 ng/mL TNF⁃αtreated rhinitis epithelial cells,P⁃AMPK protein was up⁃regulated in 10 ng/mL TNF⁃αtreated cells,the difference was statistically significant(P<0.05),but the changes of AMPK protein at different concentrations were not statistically significant(P>0.05).Compared with the normal group,the contents of TNF⁃αand IL⁃1βin the model group were increased with statistical significance(P<0.05),the apoptosis rate was decreased with statistical significance(P<0.05),and the expression level of p⁃AMPK/AMPK protein was increased with statistical significance(P<0.05).P⁃mtor/mTOR protein expression levels were significantly decreased,and the difference was statistically significant(P<0.05).Conclusion TNF⁃αinduces nasopharyngeal epithelial cell injury by activating AMPK/mTOR pathway.
