摘  要:本文旨在探索利用飞行时间质谱MALDI-TOF MS快速鉴定江苏口岸输入性病媒生物蜚蠊携带致病菌种类特征,为防控蜚蠊携带致病菌提供技术指导。通过将口岸截获的输入性病媒生物蜚蠊组织研磨后进行细菌培养,对单一菌落分别通过基质辅助激光解吸电离飞行时间质谱(Matrix-Assisted Laser Desorption/Ionization Time of Flight,MALDI-TOF MS)法进行鉴定,以检测蜚蠊携带致病菌种类特征。结果显示,通过MALDI-TOF MS法鉴定出蜚蠊携带的菌群主要是革兰阴性杆菌,包括肺炎克雷伯菌(Klebsiella pneumoniae,KPN)、产酸克雷伯菌(Klebsiella oxytoca,Kox)、铜绿假单胞菌(Pseudomonas aeruginosa,PAE)、大肠埃希菌(Escherichia coli,E.coli)、奇异变形杆菌(Proteus mirabilis)及其亚型等。少数为携带革兰阳性菌,包括金黄色葡萄球菌(Staphylococcus aureus,MRSA)及其他革兰阳性葡萄球菌的亚型、蜡样芽孢杆菌(Bacillus cereus)及其他芽孢杆菌等。综上,MALDI-TOF-MS法可实现对口岸输入性病媒生物蜚蠊携带致病菌的快速鉴定,为口岸输入性蜚蠊携带传播疾病的防控提供技术支持。This study aims to explore the rapid identification of the pathogenic bacteria carried by cockroaches,an imported vector species at Jiangsu ports,using Matrix-Assisted Laser Desorption/Ionization Time of Flight Mass Spectrometry(MALDI-TOF MS),to provide technical guidance for the prevention and control of cockroaches.By grinding the tissues of intercepted imported vector cockroaches and culturing the bacteria,we identified individual colonies using MALDI-TOF MS to detect the species characteristics of pathogenic bacteria carried by cockroaches.According to the result of MALDI-TOF MS,the main bacteria carried by cockroaches were Gram-negative bacilli,including Klebsiella pneumoniae(KPN),Klebsiella oxytoca(Kox),Pseudomonas aeruginosa(PAE),Escherichia coli(E.coli),Proteus mirabilis and their subtypes.A minority were Gram-positive bacteria,including Staphylococcus aureus(MRSA)and other Gram-positive staphylococcus subtypes,Bacillus cereus and other bacillus species.These findings indicate that MALDI-TOF-MS method can achieve rapid identification of pathogenic bacteria carried by imported vector-borne cockroaches at ports,and provide technical support for the prevention and control of imported vector-borne diseases.
