苍朴乌梅散对番泻叶致泻小鼠的作用效果研究

摘　　要：试验旨在探究苍朴乌梅散对番泻叶所致小鼠腹泻的影响。拆方试验设无苍术组、无厚朴组、全方组、番泻叶致泻模型组和空白对照组;止泻试验和肠道推进试验设置高剂量组(3.2 g/kg BW)、中剂量组(1.6 g/kg BW)、低剂量组(0.8 g/kg BW)、番泻叶致泻模型组和空白对照组。给药7 d后,采用番泻叶进行建模,以腹泻指数、血清电解质离子浓度、脏器指数和结肠病理变化为指标评价苍朴乌梅散防治腹泻的效果,肠道推进试验则灌胃炭末混悬液,计算肠道推进率。拆方试验结果显示全方组小鼠的腹泻率较低。结肠组织无损伤;止泻试验中高剂量组在2 h和6 h的止泻效果最好,中剂量组在3、4、5 h的止泻效果最好;在肠道推进试验中,各试验组肠道推进率均降低。结果表明,通过拆方试验可知全方效果较好,说明苍朴乌梅散配伍合理;通过止泻试验和肠道推进试验发现苍朴乌梅散能缓解番泻叶诱导的小鼠腹泻,改善离子失衡,保护结肠组织免受炎症细胞浸润以及涩肠等作用,推荐剂量为1.6 g/kg BW。The aim of the experiment was to inves⁃tigate the effect of Cangpu Wumei powder on senna-induced diarrhea in mice.The dismantling experiment included the no-cangzhu group,noHoupu group,full formula group,senna-induced diarrhea model group,and blank control group.The anti-diarrhea test and the intestinal propul⁃sion test were conducted with high-dose group(3.2 g/kg BW),medium-dose group(1.6 g/kg BW),low-dose group(0.8 g/kg BW),senna-induced di⁃arrhea model group,and blank control group.After seven days of administration,senna was used for mod⁃eling.The diarrhea index,serum electrolyte ion concentration,organ index and pathological changes in the colon were used as indicators to evaluate the effectiveness of Cangpu Wumei powder in preventing and treating diarrhea.The intestinal propulsion test was performed by gavage of a suspension of carbon powder,and the intestinal propulsion rate was calculated.The results of the dismantling experiment showed a lower diarrhea rate and intact colon tissue in the mice in the full formula group.In the antidiarrhea test,the high dose had the best anti-diarrhea effect at the second and sixth hours,while the me⁃dium dose had the best effect at the third,fourth,and fifth hours.Moreover,in the intestinal propulsion test,all experimental groups could reduce the rate of intestinal propulsion.The study suggests that the prescription is reasonable and the full formula has a better effect based on the dismantling experiment.Through the anti-diarrhea test and intestinal propulsion test,it is found that Cangpu Wumei powder could alleviated Senna-induced diarrhea in mice,improve ion imbalance,and protect colon tissue from inflammatory cell infiltration.The recommended dose is 1.6 g/kg BW.
