摘 要:油茶是中国重要的木本油料树种,具有较高的利用价值。本研究利用SRAP分子标记对10个福建省主栽普通油茶优良无性系进行分析,探讨其遗传差异,结果表明:从256对SRAP引物组合中筛选出12对引物对10个优良无性系进行PCR扩增,共获得186个条带,其中多态性条带166个,多态性条带的比率为89.2%。EM-2/ME-4引物组合扩增的条带多态性最高,为100%;多态性信息量最小值和最大值分别为0.280和0.394,多态性信息量平均值为0.330。10个优良无性系遗传相似系数最小值和最大值分别为0.5914和0.7742,遗传相似系数平均值为0.6708。聚类分析表明,遗传相似系数为0.6775处,10个普通油茶优良无性系可以划为4大类:闽43和闽46聚为第Ⅰ类,闽48、闽49、闽53聚为第Ⅱ类,闽50聚为第Ⅲ类,闽54、闽56、闽60、闽62聚为第Ⅳ类。这10个优良无性系之间存在着较高的遗传多样性水平,研究结果为普通油茶杂交育种和分子育种工作的开展提供参考。Camellia oleifera is an important wooden oil tree species in China,which has high value.This study used SRAP molecular markers to analysis the genetic diversity of 10 clones.The results showed that 12 pairs of primers were screened from 256 for PCR amplification.A total of 186 markers were obtained,including 166 polymorphic markers,and the polymorphism ratio of 89.2%.The polymorphism of EM-2/ME-4 primer combination was up to 100%.The minimum and maximum polymorphism information values were 0.280 and 0.394,respectively,and the average polymorphism information value was 0.330.The minimum and maximum genetic similarity coefficients of the 10 superior clones were 0.5914 and 0.7742,respectively,and the average genetic similarity coefficients were 0.6708.Cluster analysis showed that when the genetic similar coefficient was 0.6775,10 clones could be drawn to four categories.Min43 and Min46 gathered to the classⅠ,Min48,Min49,and Min53 gathered to ClassⅡ,Min50 gathered to classⅢ,and Min54,Min56,Min60 and Min62 gathered to ClassⅣ.There was a high level of genetic diversity among these 10 superior clones,and the results provided a reference for the development of hybrid breeding and molecular breeding of Camellia oleifera.
