摘　　要：目的建立高效液相色谱质谱联用仪(LC-MS)同时测定抑郁发作患者血清中西酞普兰、米氮平浓度的方法。方法设立色谱柱为Acquity UPLC BEH C_(18)柱(2.1 mm×50 mm,1.7μm),流动相为乙腈-10 mmol/L醋酸铵水溶液(均含0.1%甲酸),梯度洗脱,体积流量0.5 ml·min^(-1),柱温40℃,电喷雾电离正离子(ESI+)扫描,多反应监测模式下检测,以保留时间和离子丰度比定性,以伐地那非为内标物定量。结果西酞普兰及米氮平在各自对应的质量浓度范围内呈良好的线性关系,相关系数r^(2)>0.99,定量下限为2.5 ng/ml,相对标准偏差(RSD,n=6)为1.5%~9.3%。结论高效液相色谱质谱联用仪可同时测定抑郁发作患者血清中西酞普兰、米氮平浓度,结果精准可靠,有利于临床合理用药。Objective To establish a method for simultaneous determination of serum Citalopram and Mirtazapine in patients with depressive episode by high performance liquid chromatography-mass spectrometry(LC-MS).Methods The chromatography was performed on Acquity UPLC BEH C_(18) column(2.1 mm×50 mm,1.7μm)with mobile phase consisting of acetonitrile-10 mmol/L ammonium acetate aqueous solution(all containing 0.1%formic acid)with gradient elution at a volume flow rate of 0.5 ml·min^(-1).The column temperature was 40℃.Electrospray ionization positive ion(ESI+)scanning,multi-reaction monitoring mode detection,retention time and ion abundance ratio qualitative,vardenafil as the internal standard quantitative.Results Citalopram and Mirtazapine showed a good linear relationship in their respective mass concentration ranges,the correlation coefficient r^(2)>0.99,the lower limit of quantitation was 2.5 ng/ml,and the relative standard deviation(RSD,n=6)was 1.5%-9.3%.Conclusion High performance liquid chromatography-mass spectrometry can simultaneously determine the serum concentrations of Citalopram and Mirtazapine in patients with depression.The results are accurate and reliable,which is conducive to clinical rational drug use.
