正交试验优化铁皮石斛传统种苗繁育技术

摘　　要：为了提高铁皮石斛传统育苗腋芽萌发成苗率,指导铁皮石斛传统育苗技术在生产上的推广应用,开展影响腋芽萌发成苗率因子[繁育模式(A)、激素处理(B)、育苗基质(C)、品系(D)]的四因素五水平正交试验研究。结果显示:影响传统育苗腋芽萌发成苗率因素主次顺序为D>A>B>C,4个因素的最优水平分别是A_(2)(整条平铺并覆盖基质1 cm左右)、B_(2)(NAA 0.5 mg/L+6-BA 0.5 mg/L)、C_(1)(苔鲜)、D_(4)(紫精灵),试验筛选出传统育苗因素最优组合A_(2)B_(2)C_(1)D_(4),其萌发成苗率可达81%,较自然腋芽萌发成苗率(10%~20%)大大提高。To improve the germination rate of axillary buds in traditional Dendrobium officinale seedlings and guide the promotion and application of traditional seedling cultivation techniques in production.,a study was conducted on the factors influencing germination and seedling rates of traditional seedling-raising axillary buds.A four-factor and five-level orthogonal experiment was designed for the breeding mode(A),hormone treatment(B),seedling substrate(C),and strains(D).The results revealed that the prime and secondary order of factors affecting the germination and seedling rate of axillary buds in traditional seedling cultivation was D>A>B>C.The optimal levels of the four factors are A_(2)(the whole strip is laid flat and covered with a substrate for approximately 1 cm),B_(2)(NAA 0.5 mg·L^(-1)+6-BA 0.5 mg·L^(-1)),C_(1)(fresh moss),D_(4)(purple sprite),and the optimal combination of traditional seedling cultivation factors A_(2)B_(2)C_(1)D_(4) was selected through experiments,and its germination and seedling rates can reach 81%,which is significantly higher than the original natural axillary bud germination and seedling rates of 10%~20%.
