高效降解邻苯二甲酸酯植物内生菌的抗生素耐药性研究

摘  要:微生物降解被认为是修复邻苯二甲酸酯(PAEs)最具有前景的方法之一.然而,从环境中分离的PAEs高效降解菌大部分是致病菌或条件致病菌.植物内生菌为非致病菌,分离PAEs高效降解植物内生菌具有重要的现实意义.选取一株Bacillus subtilis N-1为研究对象,该菌株是从受污染的植物内部筛选到的植物内生菌,能以邻苯二甲酸二丁酯(DBP)和邻苯二甲酸二(2-乙基己基)酯(DEHP)两种PAEs为唯一碳源和能源生长.探究了菌株N-1的生长习性,包括其吲哚乙酸的分泌情况和对4种抗生素的耐药性.结果表明,投加色氨酸能促进菌株N-1分泌吲哚乙酸;0.50 mg·L^(-1)庆大霉素、8.00 mg·L^(-1)氯霉素、32.00 mg·L^(-1)盐酸四环素和32.00 mg·L^(-1)利福平为菌株N-1的最小抑菌质量浓度.目前污水中抗生素质量浓度远低于菌株N-1的最小抑菌质量浓度,菌株N-1修复PAEs污染的范围较广泛,具有较好的应用前景.Microbial degradation is considered as one of the most promising methods for repairing phthalates(PAEs).However,most highly efficient PAEs-degrading bacteria isolated from the environment are pathogenic or conditional pathogens.Endophytic bacteria are non-pathogenic bacteria.It is of great practical significance to isolate highly efficient PAEs-degrading endophytic bacteria.Bacillus subtilis N-1 was the research object in this study,which was an endophytic bacterium selected from a contaminated plant.Strain N-1 could grow using two types of PAEs as sole carbon source and energy source,which were dibutyl phthalate(DBP)and di(2-ethylhexyl)phthalate(DEHP).Its growth habits were explored,including the secretion of indole-3-acetic acid and the resistance to four antibiotics.The results showed that adding tryptophan could promote strain N-1 to secret indole-3-acetic acid.What's more,0.50 mg·L^(-1) gentamycin,8.00 mg·L^(-1) chloramphenicol,32.00 mg·L^(-1) tetracycline and 32.00 mg·L^(-1) rifampicin were the minimum inhibitory mass concentrations for strain N-1.At present,the concentration of antibiotics in wastewater is much lower than the minimum inhibitory mass concentration of strain N-1.Strain N-1 has a wide range of application for repairing PAEs pollution and has good prospects for application.
