摘 要:为了优化上蔡小黄姜中6-姜酚的超声提取工艺,提高6-姜酚的提取率,对影响6-姜酚提取率的提取溶剂、液料比、浸提温度、提取时间、超声功率、溶剂浓度等6个因素进行单因素试验;在单因素试验的基础上,进一步选取液料比、溶剂浓度、提取时间为影响因子,应用Box-Behnken中心组合设计建立数学模型,以6-姜酚的提取率为响应值,进行响应面分析。结果表明,超声波法提取上蔡小黄姜中6-姜酚的最优条件为:以乙醇为提取溶剂,提取时间24.12 min,液料比15.98∶1(mL∶g)和乙醇浓度62.97%。模型预测上蔡小黄姜6-姜酚提取率理论值可达到0.621%,验证值为0.619%,理论值与验证值相对误差为0.3%。本试验通过响应面法对上蔡小黄姜中6-姜酚进行提取工艺优化,可有效提升6-姜酚提取效率,为后期的开发应用提供参考依据。In order to optimize the extraction process of 6-gingerol from Dioscorea zingiberensis in Shangcai and improve the extraction yield of 6-gingerol,single factor experiments were conducted on six factors affecting the extraction rate of 6-gingerol:solvent type,liquid-solid ratio,extraction temperature,ultrasound time,ultrasound power,and solvent concentration.On the basis of single factor experiments,liquid to material ratio,solvent concentration,and ultrasound time were further selected as influencing factors to establish a mathematical model using Box-Behnken central combination design.The extraction rate of 6-gingerol was used as the response value for response surface analysis.The results showed that the optimal conditions for ultrasonic extraction of 6-gingerol from Dioscorea zingiberensis in Shangcai were as follows:ethanol as the extraction solvent,ultrasonic time of 24.12 min,liquid to material ratio of 15.98∶1(mL∶g),and ethanol concentration of 62.97%.The model predicts that the theoretical value of the extraction rate of 6-gingerol from Shangcai Xiaohuangjiang could reach 0.621%,and the verification value was 0.619%.The relative error between the theoretical value and the experimental value was 0.3%.The extraction process of 6-gingerol from Shangcai Xiaohuangjiang is optimized by response surface methodology in this experiment,which can effectively improve the extraction efficiency of 6-gingerol and provide reference for future development and application.
