地黄归芩胶囊治疗犬皮肤瘙痒症的作用机制探究

摘　　要：基于网络药理学探究地黄归芩胶囊治疗犬皮肤瘙痒症的作用机制。通过BATMAN-TCM数据库获取地黄归芩胶囊有效成分及靶点。通过GeneCards、OMIM数据库检索犬皮肤瘙痒症相关靶点,筛选出药物成分与犬皮肤瘙痒症的共有靶点,构建PPI网络,通过DAVID平台进行GO功能注释和KEGG通路富集分析。最后将预测的有效成分与核心靶点进行分子对接验证。筛选得到地黄归芩胶囊的主要活性成分有姜黄素、槲皮素、大黄素、谷氨酸、熊果酸、黄芩素等,与犬皮肤瘙痒症相关核心靶点有46个,构建的PPI网络中关联度较高的有IL-6、IL-1β、TNF等。核心靶点主要参与炎症反应、免疫反应、IL-6产生的正调控等生物过程的调节,通过作用于Th17细胞分化、JAK-STAT、TNF、PI3K-Akt等信号通路而发挥治疗作用。分子对接结果显示,药物活性成分与疾病核心靶点结合性良好。结果表明,地黄归芩胶囊中含有姜黄素、槲皮素、大黄素、熊果酸、黄芩苷等多种活性成分,主要作用于IL-6、IL-1β、TNF等靶点,可通过Th17细胞分化、JAK-STAT、TNF、PI3K-Akt等信号通路发挥治疗犬皮肤瘙痒症的作用,为进一步深入探讨其作用机制奠定了基础。To explore the mechanism of action of Dihuang Guiqin capsule in the treatment of canine skin pruritus through network pharmacology.The effective ingredients and targets of Dihuang Guiqin capsule were obtained through the BATMAN-TCM database.Targets related to canine skin itching were retrieved through GeneCards and OMIM databases,then common targets between drug components and canine skin itching were retrieved to construct a PPI network,and GO functional annotation and KEGG pathway enrichment analysis were performed using the DAVID platform.Finally,the predicted effective ingredients were validated by molecular docking with the core target.The main active ingredients screened from Dihuang Guiqin capsule including curcumin,quercetin,emodin,glutamic acid,ursolic acid,baicalin,etc.There were 46 core targets related to canine skin pruritus,and IL-6,TNF,and IL-1βwere highly correlated in the constructed PPI network.The core target mainly participated in the regulation of biological processed such as inflammatory response,immune response,and positive regulation of IL-6 production,and played a therapeutic role by regulating Th17 cell differentiation,JAK-STAT,and TNF signaling pathways.The molecular docking results showed that the active ingredients of the drug had good binding affinity with the core target of the disease.The results showed that Dihuang Guiqin capsule contain various active ingredients such as curcumin,quercetin,emodin,ursolic acid,and baicalin,mainly acting on IL-6,TNF,and IL-1βby targeting Th17 cell differentiation,JAK-STAT,and TNF signaling pathways,the therapeutic effect of treating canine skin itching can be achieved,laying a foundation for further exploring its mechanism of action.
