摘 要:【目的】分离、鉴定2022年福建省福州市某养殖场养殖的大刺鳅体表溃烂的病原,并进行药物敏感性检测,旨在为该病的防治提供参考。【方法】从患病大刺鳅体表脓肿处的脓汁中分离纯化出一株细菌,通过形态观察、生理生化特性检测及16S rDNA基因序列比对分析对该菌株进行鉴定,同时对健康大刺鳅进行人工感染试验;采用纸片扩散法检测菌株对37种药物的敏感性,并对mce1A、mig、pup、dop、whiB3、phoP、phoR 7种毒力基因进行PCR检测。【结果】从患病大刺鳅体表脓肿处分离到形态一致的菌株,将其命名为MSK20220613。经测序及Blast比对,该菌株与NCBI数据库中鰤鱼诺卡氏菌核苷酸序列的同源性高达99.86%,人工感染试验结果与自然发病的鱼体症状相似。药物敏感性检测结果显示,MSK20220613菌株对新霉素、多西环素、恩诺沙星、氟苯尼考、环丙沙星、氧氟沙星、万古霉素等21种药物敏感;对哌拉西林、青霉素、呋喃唑酮、诺氟沙星等16种药物耐受。毒力检测结果显示,MSK20220613菌株携带mce1A、pup、phoP、phoR 4种毒力基因。【结论】患病大刺鳅的病原为鰤鱼诺卡氏菌。【Objective】Drug susceptibility test was conducted on pathogens isolated from the ulcerated body surface of Mastacembelus armatus cultured in fish farm in Fuzhou,Fujian Province in 2022,so as to provide reference for the prevention and treatment of the disease.【Method】Pathogenic bacteria from sick M.armatus with skin abscess was isolated and purified,then was identified by morphological observation,physiological and biochemical characteristics and 16S rDNA sequence comparison analysis.At the same time,the artificial infection of healthy M.armatus was conducted.Furthermore,the susceptibility of the isolated strain to 37 drugs was tested by paper diffusion method,and 7 virulence genes,mce1A,mig,pup,dop,whiB3,phoP and phoR were detected by PCR.【Result】A morphologically consistent strain was isolated from the skin abscess of diseased M.armatus and named MSK20220613.Through sequencing and BLAST analysis,the strain showed a homology of 99.86%with Nocardia seriolae in the NCBI database.And the artificial infection resulted in symptoms which were similar to those of the original ones.Drug susceptibility test revealed the susceptibility of the MSK20220613 strain to 21 drugs such as neomycin,doxycycline,enrofloxacin,flufenicol,ciprofloxacin,ofloxacin,vancomycin,etc.,and resistance to 16 kinds of drugs such as piperacillin,penicillin,furazolidone,norfloxacin,etc.It carried 4 virulence genes of mce1A,pup,phoP and phoR.【Conclusion】The pathogen of M.armatus abscess disease is N.seriolae.
