摘 要:目的探索不同条件下链脲佐菌素(Streptozotocin,STZ)和高脂饮食诱导的糖尿病模型鼠血清中炎性的细胞因子、脾巨噬细胞极化表型的影响。方法选取C57BL/6J小鼠,设置对照组和实验组,对照组正常鼠粮饲养,实验组分为单纯高脂饲养组和高脂饲养联合STZ药物组,联合组给予STZ浓度为1.25、2.5和5 g/L。实验周期8周观察记录小鼠的状态、体重,测定血糖值;ELISA检测血清白细胞介素-1(Interleukin-1,IL-1)、IL-6和肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α);流式细胞术检测脾巨噬细胞CD40、CD86和CD206的表达,检测活性氧(reactive oxygen species,ROS)水平。结果与对照组相比,饮食诱导和饮食与STZ联合诱导小鼠血清IL-1、IL-6和TNF-ɑ的含量显著增高;CD40和CD86表达增加、CD206表达下降;ROS水平显著增加。结论糖尿病模型鼠的炎症性细胞因子水平升高;脾脏巨噬细胞活性氧水平增加并向M1极化。Objective To explore the effects of streptozotocin(STZ)and high-fat diet on inflammatory cytokines in serum and polariza⁃tion phenotype of splenic macrophages in diabetes model mice under different conditions.Methods C57BL/6J mice were selected and divided into the control group and the experimental group.The control group was fed with normal mouse feed,and the experimental group was divided into a pure high-fat feeding group and a high-fat feeding combined with STZ drug group.The combined group was given STZ concentrations of 1.25,2.5,and 5 g/L.The experimental period was 8 weeks,and the state,weight,and blood glucose values of the mice were observed and recorded;ELISA detection of serum interleukin-1(IL-1),IL-6,and tumor necrosis factor-α(TNF-α);Flow cytom⁃etry was used to detect the expression of CD40,CD86,and CD206 in splenic macrophages,as well as the level of reactive oxygen species(ROS).Results Compared with the control group,diet induction and diet combined with STZ significantly increased the levels of serum IL-1,IL-6,and TNF-αin mice;the expression of CD40 and CD86 increased,while the expression of CD206 decreased;The ROS level significantly increased.Conclusion The level of inflammatory cytokines in diabetes model mice is elevated;The level of reactive oxygen species in splenic macrophages increased and polarized towards M1.
