摘　　要：目的对山东省内医疗机构及血站送检的疑似ABO亚型的标本进行血清学、ABO基因测序及单倍型序列测定,分析山东省人群ABO亚型等位基因的分布和分子机制。方法血清学检测疑似为ABO亚型的无偿献血者标本239例及临床就诊患者标本701例,共940例采用PCR-SBT方法进行ABO基因序列分析;采用单克隆测序方法检测单体型;软件分析比对测序结果,确定该标本的等位基因型。结果分子测序发现山东省A基因亚型等位基因12种,以ABO*A2.05最为常见;B基因亚型等位基因15种,以ABO*BW.03最为常见;复合型ABO糖基转移酶基因6种,以ABO*BA.04最为常见;嵌合体血型6例。结论山东省B亚型的数量和种类均多于A亚型;A亚型以ABO*A2.05和ABO*A3.07最为常见;B亚型以ABO*BW.03和ABO*BW.27多见;ABO*BA.04和ABO*cisAB.01等位基因是最常见的复合型ABO糖基转移酶等位基因类型。Objective Serology,ABO gene sequencing,and haplotype sequencing were performed on samples sent to our laboratory by medical institutions and blood stations in Shandong Province for suspected ABO subgroups.The distribution and molecular mechanism of ABO subgroup alleles in the population of Shandong Province were elucidated.Methods 239 blood donors suspected to be ABO subtypes and 701 clinical patients were tested for ABO gene sequence using PCR SBT method;Using monoclonal sequencing method to detect haplotypes;The software analyzes and compares the sequencing results to determine the allele subgroup of the sample.Results Molecular sequencing revealed 12 alleles of the A gene subgroup in Shandong Province,with ABO*A2.05 being the most common;There are 15 alleles in the B gene subgroup,with ABO*BW.03 being the most common;There are 6 types of complex ABO glycosyltransferase genes,with ABO*BA.04 being the most common;6 cases of chimeric blood type.Conclusion ABO*A2.05 and ABO*A3.07 are the most common subgroups of the A gene in Shandong Province;ABO*BW.03 and ABO*BW.27 are the most common subgroups of the B gene;The ABO*BA.04 and ABO*cisAB.01 alleles are common complex ABO glycosyltransferase alleles.
