愈痫灵颗粒对癫痫大鼠Panx1/P2X7R及炎性因子表达影响的研究

摘  要:目的:研究愈痫灵颗粒对癫痫模型大鼠血清、海马组织中炎性因子IL-1β、IL-6、IL-18和TNF-α表达水平的影响,以及对大鼠海马组织Panx1/P2X7R蛋白表达的影响。方法:80只健康雄性Wistar大鼠,选择12只作为正常组,其余68只大鼠使用氯化锂-匹罗卡品腹腔注射造癫痫模型。选取造模成功的48只大鼠随机分为模型组、丙磺舒组、丙戊酸钠组和愈痫灵组,每组各12只。干预4周后,观察各组大鼠癫痫发作的频次,Nissl染色观察各组大鼠海马神经元形态,Western blot法检测各组大鼠海马组织Panx1、P2X7R蛋白表达水平,ELISA法检测各组大鼠血清及海马组织IL-1β、IL-6、IL-18和TNF-α水平。结果:与模型组比较,丙磺舒组、丙戊酸钠组、愈痫灵组大鼠癫痫发作次数减少,海马组织Panx1、P2X7R蛋白表达水平降低,血清及海马组织IL-1β、IL-6、IL-18和TNF-α水平降低,差异均有统计学意义(P<0.05)。结论:愈痫灵可降低癫痫大鼠Panx1、P2X7R蛋白表达,减少炎性因子释放。Objective:To study the effect of Yuxianling granules on the expression levels of Panx1/P2X7R protein and inflammatory factors IL-1β,IL-6,IL-18 and TNF-αin serum and hippocampus of rats with epilepsy models.Methods:Healthy male Wistar rats were selected,with 12 rats used as the normal group.The epilepsy model rats were produced by intraperitoneal injection of lithium chloride and pilocarpine.The rats in the successful model were randomly divided into model group,probenecid group,valproic acid group,and Yuxianling group,with 12 rats in each group.After 4 weeks of intervention,the frequency of epileptic seizure in rats of each group was observed.The morphology of hippocampal neurons in each group was observed by Nissl staining.The expression levels of Panx1 and P2X7R protein in the hippocampus of each group were detected by Western blot.The levels of IL-1β,IL-6,IL-18 and TNF-αin serum and hippocampus of each group were detected by ELISA.Results:Compared with the model group,the number of epileptic seizures in the probenecid group,valproic acid group,and Yuxianling group was reduced,the expression levels of Panx1 and P2X7R protein in the hippocampus of rats were reduced,and the levels of IL-1β,IL-6,IL-18 and TNF-αin serum and hippocampus of rats were also reduced,the differences were statistically significant(P<0.05).Conclusion:Yuxianling can reduce the expression of Panx1 and P2X7R protein in epileptic rats and reduce the release of inflammatory factors.
