“标本配穴”电针内关、足三里对慢性心肌缺血大鼠炎性损伤的影响

摘  要:目的:观察“标本配穴”电针治疗对慢性心肌缺血大鼠炎性损伤的影响。方法:21只雄性SD大鼠随机分为对照组、模型组、电针组,每组各7只。模型组、电针组连续7 d皮下多点位注射盐酸异丙肾上腺素[5 mg(/kg·d)]制备慢性心肌缺血大鼠模型。造模成功后,电针组大鼠电针内关、足三里21 d,1次/d,20 min/次;对照组和模型组大鼠常规饲养,同等抓取,不予干预。采用PowerLab多导生理记录仪记录分析3组大鼠造模前、造模后、干预后心电图的变化,ELISA法检测大鼠心肌组织中炎症因子IL-17、IL-10的含量,实时荧光定量PCR技术检测3组大鼠心肌组织中JAK1、STAT3 mRNA表达水平。结果:造模后,模型组大鼠ST段高度变化值明显高于对照组,差异有统计学意义(P<0.05),表明造模成功;干预21 d后电针组大鼠ST段高度变化值(干预后-造模后)低于模型组,差异有统计学意义(P<0.05)。与对照组比较,模型组大鼠心肌组织中IL-17含量及JAK1、STAT3 mRNA相对表达量均升高,IL-10含量减少,差异均有统计学意义(P<0.05);与模型组比较,电针组大鼠心肌组织中IL-17含量及JAK1、STAT3 mRNA相对表达量均降低,IL-10含量升高,差异均有统计学意义(P<0.05)。结论:“标本配穴”电针可能通过JAK1/STAT3信号通路调节IL-17、IL-10炎症因子表达而减轻炎性损伤,改善心功能。Objective:To observe the effects of electroacupuncture(EA)with“Biao-Ben acupoints combination”on inflammatory injury in rats with chronic myocardial ischemia.Methods:Twenty-one Sprague-Dawley(SD)rats were randomly divided into a control group,a model group,and an EA group,with seven rats in each group.The model group and the EA group were injected subcutaneously with multiple doses of isoproterenol hydrochloride[5 mg(/kg·d)]for seven consecutive days to establish a chronic myocardial ischemia rat model.After successful modeling,the rats in the EA group received EA at Neiguan and Zusanli for 21 days,once daily for 20 minutes.The control group and the model group were conventionally raised and handled equally without any intervention.The changes in the electrocardiogram(ECG)before and after modeling and after intervention were recorded and analyzed using a PowerLab multichannel physiological recorder.The levels of inflammatory factors interleukin-17(IL-17)and interleukin-10(IL-10)in the heart tissue were detected using the enzyme-linked immunosorbent assay(ELISA)method.The expression levels of JAK1 and STAT3 mRNA in the myocardial tissues of the three groups were measured using real-time fluorescent quantitative PCR technology.Results:After modeling,the ST segment elevation change value in the model group was significantly higher than that in the control group with a statistically significant difference(P<0.05),indicating successful modeling.After 21 days of intervention,the ST segment height change value(post-intervention-post-modelling)in the EA group was lower than that in the model group,with a statistically significant difference(P<0.05).Compared with the control group,the levels of IL-17 and the relative mRNA expression of JAK1 and STAT3 were increased,and the level of IL-10 was decreased in the myocardial tissues of the model group,with statistically significant differences(P<0.05).Compared with the model group,the levels of IL-17 and the relative mRNA expression of JAK1 and STAT3 were decreased,and the
