摘 要:目的:分析太子参环肽的主要入血成分。方法:SPF级雄性SD大鼠灌胃太子参环肽提取物,不同时间点于眼眶取血。离心分离血浆样品,通过超高效液相色谱-飞行时间质谱(UPLC-Q-TOF-MS)技术进行分析,采用Waters ACQUITY UPLC■CSH C18(2.1×100 mm,1.7μm)色谱柱;以乙腈、0.1%甲酸水溶液为流动相,梯度洗脱。结合分子离子和碎片离子特征峰信息,分析不同环肽在大鼠体内的入血原型成分及代谢物。结果:经液质联用分析,共检测出15个入血成分,其中有7个为原型成分,8个为代谢产物。代谢途径主要包括Ⅰ相的水解、脱甲基戊羧酸、脱氢、加氢和Ⅱ相的甲基化结合反应。结论:初步鉴定了太子参环肽的入血原型成分及代谢产物,为其进一步的药效物质基础及质量控制研究提供了数据支撑。Objective:To analyze the major blood-entry components of Heterophyllin.Methods:Male SPF-grade SD rats were orally administered Heterophyllin extract,and blood samples were collected from the orbital sinus at different time points.Plasma samples were separated by centrifugation and analyzed using ultra-performance liquid chromatography coupled with quadrupole-time-of-flight mass spectrometry(UPLC-Q-TOF-MS).A Waters ACQUITY UPLC®CSH C18(2.1×100 mm,1.7μm)column was used,with acetonitrile and 0.1%aqueous formic acid as the mobile phases under gradient elution.Based on the molecular ions and fragment ion characteristic peaks,the prototype components in blood and metabolites of different cyclotides in the rat were analyzed.Results:After oral administration of Heterophyllin extract to rats,a total of 15 blood-entry components were detected by LC-MS analysis,including 7 prototype components and 8 metabolites.The metabolic pathways mainly included phase I hydrolysis,demethylation to carboxylic acid,dehydrogenation,hydrogenation,and phase II methylation conjugation reactions.Conclusion:The blood-entry prototype components and metabolites of Heterophyllin were preliminarily identified,providing data support for further studies on the pharmacological material basis and quality control.
