摘  要:目的:探讨氧化苦参碱对人卵巢癌CAOV3细胞的生长抑制作用及机制。方法:体外培养人卵巢上皮浆液性癌细胞系CAOV3细胞,氧化苦参碱给药剂量按照0 mg/ml,2 mg/ml,4 mg/ml,8 mg/ml作用于CAOV3细胞24 h,48 h,采用噻唑蓝(MTT)法检测细胞增殖能力,筛选药物最佳作用浓度与药物作用时间;选用4 mg/ml的氧化苦参碱溶液作用于CAOV3细胞,采用划痕实验,检测细胞迁移率;采用Western blotting法检测Caspase3、Bax以及Bcl-2蛋白表达水平;采用流式细胞术检测细胞凋亡率。结果:与空白组相比,氧化苦参碱可以明显降低CAOV3细胞的细胞活力,且抑制作用具有时间依赖性及剂量依赖性,差异具有统计学意义(P<0.05);同时筛选出4 mg/ml氧化苦参碱作用于CAOV3细胞48 h为最佳有效条件。与空白组相比,氧化苦参碱可以抑制CAOV3细胞的迁移,且作用具有时间依赖性,差异具有统计学意义(P<0.05);氧化苦参碱作用于CAOV3细胞后明显升高Caspase3以及Bax蛋白的表达,明显降低Bcl-2蛋白的表达,差异具有统计学意义(P<0.05);氧化苦参碱可以促进CAOV3细胞的早期凋亡。结论:氧化苦参碱可能通过促进细胞的早期凋亡来抑制CAOV3细胞的生长以及迁移,其作用可能与促进Caspase3、Bax蛋白表达、抑制Bcl-2蛋白表达有关。Objective:To investigate the growth inhibitory effect of oxymatrine on human ovarian cancer CAOV3 cells and its underlying mechanism.Methods:Human ovarian serous epithelial cancer cell line CAOV3 cells were cultured in vi-tro.Oxymatrine was applied at concentrations of 0 mg/ml,2 mg/ml,4 mg/ml,and 8 mg/ml for 24 hours and 48 hours.The cell proliferation capacity was detected using the thiazolyl blue(MTT)method to determine the optimal concentration and duration of drug action.Scratch assays were conducted using 4 mg/ml oxymatrine to evaluate cell migration.Western blotting was used to measure the protein levels of Caspase3,Bax,and Bcl-2.Flow cytometry was employed to assess the cell apop-tosis rate.Results:Compared to the blank group,oxymatrine significantly reduced the viability of CAOV3 cells,with the inhibitory effect being time-and dose-dependent,with statistically significant differences(P<0.05).The optimal effective condition was determined to be 4 mg/ml oxymatrine for 48 hours.Compared to the blank group,oxymatrine inhibited the mi-gration of CAOV3 cells,with the effect being time-dependent,and the difference was statistically significant(P<0.05).Oxymatrine increased the expression of Caspase3 and Bax proteins and decreased the expression of Bcl-2 protein in CAOV3 cells,and the difference was statistically significant(P<0.05).Oxymatrine promoted early apoptosis in CAOV3 cells.Con-clusion:Oxymatrine may inhibit the growth and migration of CAOV3 cells by promoting early apoptosis,and its mechanism may involve promotion of Caspase3 and Bax protein expression and inhibition of Bcl-2 protein expression.
