摘 要:本试验利用生物反应器对ST细胞全悬浮培养参数以及猪瘟病毒的繁殖工艺进行探索。从培养基、细胞接种浓度、病毒接毒量三个方面进行工艺优化,放大培养,建立了猪瘟病毒的全悬浮培养工艺。结果表明:当细胞接种浓度为1.0×10^(6)/mL、病毒接毒MOI为0.05、采用培养基1进行培养时,病毒含量达到10^(7.5)FAID_(50)/mL,工艺验证两次,结果都符合要求,稳定可靠,为猪瘟疫苗大规模全悬浮培养奠定了基础。This experiment explored the parameters of ST cell suspension culture and the reproductive process of classical swine fever virus using a bioreactor.The process was optimized from the culture medium,cells inoculation concentration and viral vaccination dose,and the whole suspension culture process of classical swine fever virus was established.The results showed that when the cell inoculation concentration was 1×106/mL and the MOI of virus inoculation was 0.05,and the medium 1 was used,the viral titer reached 107.5FAID50/mL.The process was validated twice,and the results met the requirements,which were stable and reliable,laying the foundation for large-scale suspension culture of classical swine fever vaccine industrialization.
