摘  要:本试验旨在探讨使用微载体生物反应器培养猪传染性胃肠炎病毒(Transmissible gastroenteritis virus,TGEV)的效果,对比不同时间段培养出的抗原效价,从而确定TGEV培养的最佳收获时间,提升抗原效价,为最终生产出高质量、品质稳定的TGEV活疫苗奠定基础。在生物反应器细胞上罐时每次接入细胞量达到1.2×10~9个,按相同的条件进行培养。当微载体上的细胞长至单层致密时,进行细胞接毒,接毒后4 h开始取样,接着每隔2 h留样,18 h后每隔1 h取样1次,直到载体上细胞总脱落面积达80%~85%结束取样,检测抗原效价。结果表明:在接毒后20~21 h病毒效价达到峰值,故确定在接毒后20~21 h为病毒最佳收获时间。The purpose of this study was to investigate the effect of using microcarrier bioreactor to culture transmissible gastroenteritis virus(TGEV),and to compare the antigen titers cultured at different time periods,so as to determine the optimal harvest time of TGEV culture,improve the antigen titer,and lay the foundation for the final production of high-quality and stable live TGEV vaccine.The amount of cells reached 1.2×109 each time when the bioreactor cells were put into the tank,and the cells were cultured under the same conditions.When the cells on the microcarrier grew to a dense monolayer,the cells were inoculated with the virus,and the samples were taken at 4 hours(h)after inoculation,then the samples were retained every 2 h,and the samples were taken every 1 h after 18 h,the sampling was finished until the total shedding area of the cells on the carrier reached 80%~85%,then the antigen titer were detected.The results showed that the virus titer reached the peak at 20 h to 21 h after inoculation,so it was determined that 20~21 h after inoculation was the best harvest time for the virus.
