摘 要:建立高效液相色谱法同时测定晕可平糖浆中5-羟甲基糠醛、丹参素、原儿茶醛、咖啡酸、苯甲酸、毛蕊花糖苷、异毛蕊花糖苷、迷迭香酸、甘草酸9种成分的含量。采用Phenomenex luna C_(18)色谱柱(250 mm×4.6 mm,5μm),以乙腈-0.05%三氟乙酸溶液为流动相进行梯度洗脱,流量为1 mL/min,柱温为25℃,检测波长分别为245 nm(苯甲酸、甘草酸)、280 nm(5-羟甲基糠醛、丹参素、原儿茶醛)和330 nm(咖啡酸、毛蕊花糖苷、异毛蕊花糖苷、迷迭香酸)。9种成分的质量浓度在各自范围内与对应的色谱峰面积线性关系良好,相关系数均为0.9999,方法检出限为0.01~0.21μg/mL,定量限为0.02~0.68μg/mL。样品加标平均回收率为96.9%~105.0%,测定结果的相对标准偏差为1.3%~3.4%(n=6)。该方法简便可靠,重复性好,可用于晕可平糖浆的质量控制。A method for simultaneous determination of 5-hydroxymethylfurfural,danshensu,protocatechuic aldehyde,caffeic acid,benzoic acid,acteoside,isoacteoside,rosmarinic acid and glycyrrhizic acid in Yunkeping syrup by high performance liquid chromatography was established.Phenomenex luna C_(18) column(250 mm×4.6 mm,5μm)was used with the mobile phase comprising of acetonitrile-0.05%trifluoroacetic acid at a flow rate of 1.0 mL/min in a gradient elution manner.The column temperature was 25℃,and the detection wavelengths were 245 nm for benzoic acid and glycyrrhizic acid,280 nm for 5-hydroxymethylfurfural,danshensu and protocatechuic aldehyde,330 nm for caffeic acid,acteoside,isoacteoside and rosmarinic acid,respectively.The mass concentrations of the 9 components had a good linear relationship with the corresponding chromatographic peak areas in their respective ranges,with correlation coefficients of 0.9999.The detection limits were 0.01-0.21μg/mL,and the quantification limits were 0.02-0.68μg/mL.The average recoveries of samples spiked were 96.9%-105.0%,and the relative standard deviations of the determination results were 1.3%-3.4%(n=6).The method is simple,reliable and reproducible,which can be used for the quality control of Yunkeping syrup.
