高效液相色谱法测定枸杞中对香豆酸

摘  要:建立高效液相色谱法测定枸杞中对香豆酸的含量。采用Waters Atlantis T3色谱柱(250 mm×4.6 mm,5μm)为分离柱,以乙腈-0.1%磷酸(体积比为13∶87)为流动相,流量为1.0 mL/min,进样体积为10μL,柱温为30℃,检测波长为310 nm。对香豆酸的质量浓度在0.28~13.78 mg/L范围内与色谱峰面积线性关系良好,线性相关系数为0.9999,方法检出限为2.03 mg/kg,定量限为6.06 mg/kg。测定结果的相对标准偏差为0.62%(n=6),样品加标平均回收率为97.97%。该方法的精密度和准确度良好,可用于枸杞中对香豆酸含量的测定。A method for the determination of p-coumaric acid in Lycium barbarum by high performance liquid chromatography was established.Waters Atlantis T3 column(250 mm×4.6 mm,5μm)was used as the separation column.The mobile phase was acetonitrile-0.1%phosphoric acid(the volume ratio was 13∶87),the flow rate was 1.0 mL/min,the injection volume was 10μL,the column temperature was 30℃,and the detection wavelength was 310 nm.The mass concentration of p-coumaric acid had a good linear relationship with the chromatographic peak area in the range of 0.28-13.78 mg/L,the linear correlation coefficient was 0.9999.The detection limit of the method was 2.03 mg/kg,and the quantitative limit was 6.06 mg/kg.The relative standard deviation of the determination results was 0.62%(n=6),and the average recovery rate of the sample was 97.97%.This method has good precision and accuracy,and which can be used for the determination of p-coumaric acid in Lycium barbarum.
