响应面法探究纳豆激酶活性保留条件及肠溶性胶囊的保护作用

摘  要:采用纤维蛋白降解法测定活性,响应面法分析纳豆激酶在不同因素中活性变化,明确纳豆激酶最优活性保留条件。采用胃液-肠液串联式方法,比较普通纳豆激酶胶囊与肠溶性胶囊在人工胃肠液中的稳定性,为开发肠溶性纳豆激酶胶囊做理论支撑。结果表明纳豆激酶活性保留最优条件为:PH=8、转速为40 r/min、时间为1.3 h、温度为23℃。此时纳豆激酶活性保留为20291.7 FU/g。在人工胃液处理后,活性下降到30.9%。普通胶囊与肠溶性胶囊在人工胃肠液处理后活性分别为28.0%、53.1%。最终结论为纳豆激酶在酸性的胃液中极不稳定,活性丧失巨大。在人工肠液中酶较稳定。肠溶性胶囊与普通胶囊相比具有保护作用,将纳豆激酶制成肠溶性的产品服用,能使得纳豆激酶更加稳定、活性更高。The activity of nattokinase was determined using fibrin degradation method.Response surface methodology was employed to analyze the changes in nattokinase activity under different factors,aiming to identify the optimal conditions for preserving its activity.An artificial gastric-intestinal fluid tandem method was used to compare the stability of regular nattokinase capsules and enteric-coated capsules in artificial gastrointestinal fluid,providing theoretical support for the development of enteric-coated nattokinase capsules.The results indicated that the optimal conditions for preserving nattokinase activity were pH=8,rotation speed of 40 r/min,1.3 hours,and at 23℃.Under these conditions,the preserved nattokinase activity was 20291.7 FU/g.After artificial gastric fluid treatment,the activity decreased to 30.9%.The activity of regular capsules and enteric-coated capsules after artificial gastric fluid treatment was 28.0%and 53.1%,respectively.The final conclusion was that nattokinase was extremely unstable in acidic gastric fluid with a huge loss of activity.In artificial intestinal fluid the enzyme was more stable.Enteric soluble capsules had a protective effect compared to regular capsules,and taking nattokinase as an enteric soluble product could make nattokinase more stable and active.
