摘 要:为探究宽体金线蛭、天津医蛭和菲牛蛭等3种药用水蛭的染色体制备及核型分析方法,丰富对水蛭染色体数目及核型组成的认知,以精巢细胞为材料,采用空气干燥法制作前述3种水蛭的染色体玻片,并进行镜检和核型分析。试验结果显示,3种水蛭的染色体数目和核型公式分别为宽体金线蛭:2n=22=8m+4sm+4st+6t,天津医蛭:2n=22=12m+4sm+2st+4t,菲牛蛭:2n=26=4m+4sm+6st+12t;核型不对称系数分别为宽体金线蛭71.27%,天津医蛭65.70%,菲牛蛭78.99%;核型分类标准分别为宽体金线蛭“3A”型,天津医蛭“2A”型,菲牛蛭“3A”型。结果说明,核型分析结果与这3种水蛭所在的系统分类地位相符,3种水蛭的进化程度有一定差别。To investigate the methods of chromosome preparation and karyotype analysis of three medicinal leech species,Whitmania pigra,Hirudo tianjinensis,and Poecilobdella manillensis,and to further enrich the knowledge of the number of chromosomes and karyotype composition of leeches.The spermatocytes were used as material to make chromosome slides of the three leech species by air-drying method,and microscopic examination and karyotype analysis were carried out.The results showed that the chromosome number and karyotype formulas of the three leech species were as follows:W.pigra:2n=22=8m+4sm+4st+6t,H.tianjinensis:2n=22=12m+4sm+2st+4t,P.manillensis:2n=26=4m+4sm+6st+12t.Karyotype asymmetry coefficients were 71.27%,65.70%and 78.99%,respectively.The karyotype classification criteria were the“3A”type of W.pigra,“2A”type of H.tianjinensis and“3A”type of the P.manillensis.The results indicated that the karyotype analysis was consistent with the phylogenetic status of the three leech species,which was different in their degree of evolution to some extent.
