摘　　要：目的建立苯甲酸琥珀酰亚胺酯衍生化法测定复方甘草酸苷制剂中常量组分氨基酸。方法以苯甲酸琥珀酰亚胺酯为衍生试剂,采用Angla Innoval C_(18)色谱柱(150 mm×4.6 mm,5μm)分离,流动相为甲醇-体积分数0.5%三乙胺磷酸缓冲液(pH 5.00)进行梯度洗脱,检测波长为236 nm,进样量为10μL,柱温为室温,流速为1.0 mL·min^(-1)。结果复方甘草酸苷制剂中甘氨酸、蛋氨酸的质量浓度分别在0.501~50.1 mg·L^(-1)、1.00~50.2 mg·L^(-1)内线性关系良好,相关系数(r)分别为0.9996、0.9997;回收率满足制剂分析中常量组分分析要求。结论该方法减小了高灵敏度衍生试剂分析常量组分需多次稀释带来的操作误差,且过量的衍生试剂水解后不干扰分离,该方法可作为药物制剂中常量氨基酸的分析方法。Objective To establish a method for the analysis of amino acids in Compound Glycyrrhizin preparations derived from succinimide benzoate.Methods Succinimide benzoate was used as the derivatization reagent.The separation was performed on Angla innovation C_(18)column(150 mm×4.6 mm,5μm),methanol as mobile phase A,0.5%triethylamine phosphate buffer(pH 5.00)as mobile phase B with gradient elution.The detection wavelength was 236 nm,the injection volume was 10μL,the column temperature was room temperature,and the flow rate was 1.0 mL·min^(-1).Results Glycine and methionine in Compound Glycyrrhizin preparation had the good linear relationship in the range of 0.501-50.1 mg·L^(-1)and 1.00-50.2 mg·L^(-1)respectively,r=0.9996,r=0.9997;The recovery rate met the requirements of content component analysis in preparation analysis.Conclusion The method reduces the operation error caused by multiple dilution of content components in the analysis of high-sensitivity derivative reagents,but also does not interfere with the separation after hydrolysis of excessive derivative reagents.The method is simple,fast and accurate.It can be used as an analysis method of constant amino acids in pharmaceutical preparations.
