摘 要:为探究中药多糖(TCMP)对自然衰老小鼠卵巢功能的延缓作用,本试验将40只10月龄C57小鼠随机分为4个组,每组10只。对照组(Control)每天灌服0.2 mL去离子水,枸杞多糖组(LBP)、菟丝子多糖组(CCP)和熟地黄多糖组(PRRP)分别按200 mg/(kg·bw·d)剂量灌服枸杞多糖、菟丝子多糖和熟地黄多糖。用药30 d,每天观察小鼠基本情况,并拍照记录。采用Morris水迷宫试验和跳台试验检测TCMP对小鼠潜伏期的影响;眼球采血检测小鼠血清中超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、谷胱甘肽过氧化物酶(GSH-Px)、总抗氧化能力(T-AOC)和丙二醛(MDA)水平;称取小鼠体质量,处死小鼠后收集肾脏、卵巢和子宫,计算脏器指数;苏木精-伊红(H.E.)染色观察卵巢组织形态学变化;流式细胞术检测小鼠脾脏组织中调节性T细胞(Treg)的比例。结果显示,与对照组相比,中药多糖处理组小鼠的各项指标均有所改善;菟丝子多糖组小鼠在Morris水迷宫试验中的逃避潜伏期缩短,但差异没有统计学意义(P>0.05),跳台试验中测试潜伏期和错误次数均极显著降低(P<0.01)。相较于枸杞多糖和熟地黄多糖,菟丝子多糖对小鼠脱毛程度、体质量和脏器指数的改善效果更为显著;菟丝子多糖还能够显著提高血清中SOD、CAT、GSH-Px和T-AOC水平(P<0.05或P<0.01),极显著降低MDA含量(P<0.01),增加卵巢中各类卵泡的数量,极显著降低CD4^(+)CD25^(+)Foxp3^(+)Treg细胞的比例(P<0.01)。结果表明,3种多糖能在一定程度上缓解小鼠卵巢衰老,菟丝子多糖呈现出比枸杞多糖和熟地黄多糖更为积极的效果。本试验结果可为开发改善卵巢衰老的药物提供参考依据。To investigate the delaying effects of traditional Chinese medicine polysaccharides(TCMP) on ovarian function in naturally aging mice,this study divided 40 ten-month-old C57 mice into four groups,with 10 mice in each group.The control group(Control) was orally administered 0.2 mL of deionized water daily,while the Lycium barbarum polysaccharide group(LBP),Cuscuta chinensis polysaccharide group(CCP),and Rehmannia glutinosa polysaccharide group(PRRP) were respectively administered 200 mg/(kg·bw·d) of Lycium barbarum polysaccharide,Cuscuta chinensis polysaccharide,and Rehmannia glutinosa polysaccharide.After 30 days of treatment,the general conditions of the mice were observed daily and recorded through photographs.The effects of TCMP on the latency period of the mice were assessed using the Morris water maze and step-down tests.Blood samples were collected from the eyes to measure serum levels of superoxide dismutase(SOD),catalase(CAT),glutathione peroxidase(GSH-Px),total antioxidant capacity(T-AOC),and malondialdehyde(MDA).Body weights of the mice were recorded,and kidneys,ovaries,and uteri were collected post-mortem to calculate organ indices.Hematoxylin and eosin(H.E.) staining was used to observe histomorphological changes in ovarian tissue,and flow cytometry was employed to detect the proportion of regulatory T cells(Treg) in the spleen tissue of the mice.The results showed that,compared to Control group,various indicators in the TCMP-treated groups improved.The Cuscuta chinensis polysaccharide group exhibited a shorter escape latency in the Morris water maze test,though the difference was not statistically significant( P > 0.05).However,in the step-down test,both the test latency and error times were significantly reduced( P < 0.01).Compared to Lycium barbarum polysaccharide and Rehmannia glutinosa polysaccharide,Cuscuta chinensis polysaccharide more effectively improved hair loss,body weight,and organ indices.Cuscuta chinensis polysaccharide also significantly increased serum levels of SOD,CAT,GSH-Px,and T
