摘 要:为研究黄芪提取物中抗氧化活性成分及作用机制,使用石油醚、乙酸乙酯、氯仿和正丁醇4种有机试剂分别提取黄芪的有效成分,并通过高效液相色谱(high performance liquid chromatography,HPLC)法进行检测。基于网络药理学研究其抗氧化效果,采用网络药理学多个线上数据库获取黄芪—抗氧化共有靶点并构建蛋白互作网络(protein⁃protein interaction networks,PPI)以及进行基因本体(gene ontology,GO)富集分析和京都基因与基因组百科全书(Kyoto Encyclopedia of Genes and Genomes,KEGG)信号通路富集分析。结果表明:黄芪各有机相提取物对·OH、O2-·和DPPH·均有一定的清除能力,黄芪乙酸乙酯相提取物和黄芪氯仿相提取物的清除效果较好,主要的抗氧化活性成分为山奈酚、齐墩果酸和没食子酸。通过线上数据库筛选出黄芪抗氧化靶点136个。GO富集表明这些靶点大多与无机物反应、激素反应、对外来刺激反应有关,KEGG富集分析显示这些靶点与癌症通路、脂质与动脉粥状硬化、缺氧诱导因子信号通路有关。分子对接结果显示,所提取的活性成分抗氧化效果较好。To investigate the antioxidant active components and mechanisms of Astragalus root extract,effec⁃tive components were extracted using four organic solvents:petroleum ether,ethyl acetate,chloroform,and n⁃butanol and detected using high performance liquid chromatography(HPLC).The antioxidant effects were studied based on network pharmacology.Multiple online databases were employed to identify common targets between Astragalus and antioxidants.Protein⁃protein interaction networks(PPI)were constructed,and gene ontology(GO)enrichment analysis and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway enrich⁃ment analysis were performed.The results showed that all Astragalus root extracts had certain scavenging abili⁃ties against·OH,O2-·,and DPPH·radicals.The ethyl acetate and chloroform extracts of Astragalus exhibited better scavenging effects,with kaempferol,oleanolic acid,and gallic acid identified as the main antioxidant ac⁃tive components.A total of 136 antioxidant targets for Astragalus were screened through online databases.GO enrichment indicated that these targets were mostly related to responses to inorganic substances,hormones,and external stimuli.KEGG enrichment analysis revealed that these targets were associated with cancer path⁃ways,lipid and atherosclerosis,and the hypoxia⁃inducible factor signaling pathway.The molecular docking re⁃sults showed better antioxidant effect of the extracted active ingredients.
